电针干预对大鼠骨密度及骨髓间充质干细胞成脂分化的影响

【摘要】 目的探讨电针治疗绝经后骨质疏松症的可能机制。方法选取3月龄健康雌性SD大鼠48只,随机分为正常组、模型组、雌激素组、电针组,每组12只。通过双侧卵巢切除法模拟绝经后骨质疏松大鼠模型。造模成功后,雌激素组给予尼尔雌醇1次/周灌胃,连续给药12w;电针组电针肾俞、脾俞穴、关元、足三里、三阴交等穴,每日一次,十次一疗程,疗程间间隔3d,共治疗7个疗程。治疗结束后,采用双能X线骨密度仪检测右侧股骨骨密度,通过全骨髓贴壁法分离BMSCs并进行原代和传代培养,传至3代后进行成脂诱导,RTPCR法检测LPL-mRNA的表达,观察BMSCs成脂的分化。结果模型组骨密度值明显低于正常组(P<0.01),雌激素组与电针组的骨密度值高于模型组(P<0.01),雌激素组与电针组之间比较差异无统计学意义(P>0.05);成脂诱导BMSCs后,模型组LPL-mRNA表达高于正常组(P<0.01),雌激素组和电针组LPL-mRNA表达低于模型组(P<0.01),雌激素组和电针组比较,差异无统计学意义(P>0.05)。结论电针干预对骨质疏松的治疗作用,可能与抑制BMSCs成脂分化有关。

【Abstract】 Objective To explore the possible mechanism of electroacupuncture in the treatment of postmenopausal osteoporosis. Methods 48 healthy female SD ratsaged 3 months were selected and randomly divided into normal group, modelgroup, estrogen group and electroacupuncture group, with 12 rats in each group. Rats with postmenopausal osteoporosis were simulated by bilateral ovariectomy. After the model was successfully established, the estrogen group was given nylestradiol, which was given by gavage once per week for 12 w continuously. Inthe electroacupuncture group, shenshu, pi shu,guan yuan, zusanli sanyin jiao and so on,which were treated once a day with a course of treatment of 10 times, with a 3 d interval between the courses of treatment, and a total of 7 courses of treatment. After the treatment, dual-energy X-ray absorptiometry was used to detect the bone mineral density of the right femur. BMSCs were isolated by whole-bone marrow adherent method and were cultured in the primary and subculture. Lipid induction was conducted after the transfer to the third generation, and lpl-mrna expression was detected by rt-pcr to observe the lipid differentiation of BMSCs. Results BMD of model group was significantly lower than that of normal group(P< 0.01), BMD of estrogen group and electroacupuncture group was higher than that of model group(P< 0.01), and there was no statistically significant difference between estrogen group and electroacupuncture group(P>0.05). After the induction of BMSCs by lipid formation, the expression of lpl-mrna in the model group was higher than that in the normal group(P< 0.01), and the expression of lpl-mrna in the estrogen group and electroacupuncture group was lower than that in the model group(P< 0.05). There was no statistically significant difference between the estrogen group and the electroacupuncture group(P>0.05). Conclusion the effect of electroacupuncture on osteoporosis may be related to the inhibition of BMSCs lipid differentiation.
