玉米须提取物对糖尿病足小鼠血管内皮相关因子及炎症因子的影响
Effects of Corn Silk Extract on Vascular Endothelial Related Factor and Inflammatory Factor in Diabetic Foot Mice
【摘要】 目的:探讨玉米须提取物对糖尿病足小鼠模型血管内皮相关因子、炎症因子的影响。方法:昆明种小鼠150只,适应性饲养1周,随机分成5组,30只/组,分别为空白组、模型对照组、玉米须提取液高剂量组、中剂量组、低剂量组。空白组喂以标准饲料,其余组喂以高脂高糖饮食至实验结束。11周后除空白组外均造模,造模前禁食12 h,自由饮水,按每天40 mg/kg体重剂量,将链脲佐菌素(STZ)用浓度为0.1 mmol/L柠檬酸缓冲液(pH4.2)溶解,给小鼠做腹腔注射1次/d,连续5 d。5 d后,测量小鼠空腹6 h后血糖值,大于11.1 mmol/L者视为糖尿病模型造模成功,然后采用结扎小鼠股动脉形成肢体局部缺血的糖尿病足小鼠模型。除空白组和模型对照组外,其余3组玉米须提取物按照400、200、100 mg/kg,配成浓度为20、10、5 g/L,以0.2 mL/10 g灌胃给药。给药第30天、给药后1 h、禁食12 h,小鼠尾部取血,观察小鼠用药前后的血糖、血管内皮相关因子、炎症因子的指标变化。结果:给药后玉米须提取物高、中、低剂量组与模型对照组相比较均具有降低糖尿病足小鼠模型血糖的作用(P<0.01),且呈现剂量依赖性,高剂量组要明显优于低剂量组的降糖效果;给药后高、中、低剂量组与模型对照组相比较均具有降低VEGF、ET-1和提升NO的作用(P<0.01),且呈现剂量依赖性,高剂量组要明显优于低剂量组降低VEGF、ET-1和提升NO的作用;给药后高、中、低剂量组与模型对照组相比较均显著降低CRP、IL-6、TNF-α的作用(P<0.01),且呈现剂量依赖性,高剂量组要明显优于低剂量组(P<0.05)。结论:玉米须提取物具有有效改善小鼠糖尿病足模型血糖、血管内皮相关因子、炎症因子的作用,且改善的效果随着剂量浓度的增高而增加。
【Abstract】 Objective To investigate the effects of corn silk extract on vascular endothelial related factors and inflammatory factors in diabetic foot mice. Methods 150 Kunming mice were randomly divided into 5 groups, 30 mice/group, which were blank group, model control group, high-dose group of corn silk extract, medium-dose group and low-dose group. The blank group was fed with standard diet, and the other groups were fed with high-fat and high-sugar diet until the end of the experiment. After 11 weeks, STZ was dissolved in 0.1 mmol/L citric acid buffer(pH4.2) at a daily dose of 40 mg/kg body weight. The mice were given intraperitoneal injection once a day for 5 days. Five days later, the diabetic foot model was established successfully when the blood glucose value was greater than 11.1 mmol/L 6 hours after fasting. Then the diabetic foot model with local limb ischemia was established by ligating the femoral artery of the mice. Except the blank group and model control group, the other three groups of corn silk extracts were administered by gavage at the concentration of 20, 10 and 5 g/L and the volume of gavage was 0.2 ml/10 g according to 400, 200 and 100 mg/kg. On the 30 th day, 1 h after administration, 12 h after fasting, blood samples were taken from the tail of mice to observe the changes of blood sugar, vascular endothelium-related factors and inflammatory factors before and after ad ministration. Results Compared with the model control group, the high, middle and low dose groups of corn extract had the effect of reducing the blood sugar of diabetic foot mice model(P < 0.01), and showed a dose-dependent, there was a difference between the groups(P < 0.05), the high dose group was significantly better than the low dose group in reducing the blood sugar effect. Compared with the model control group, the high, middle and low dose groups had the effect of decreasing VEGF, ET-1 and increasing NO(P < 0.01), and showed a dose-dependent, there was a difference between the groups(P < 0.05). The high dose group was significantly better than the low dose group in decreasing VEGF, ET-1 and increasing No; After administration, the effects of CRP, IL-6 and TNF-α in the high, middle and low dose groups were significantly lower than those in the model control group(P < 0.01), and showed a dose-dependent, there was a difference between the groups, the high dose group was significantly better than the low dose group(P < 0. 05). Conclusion Corn silk extract can effectively improve the effects of blood sugar, vascular endothelial related factors and inflammatory factors in diabetic foot model of mice, and the improvement effect increases with the increase of dose concentration.
