植物乳杆菌共培养诱导产细菌素基因座的遗传分析
Heredity Analysis of the Plantaricin Locus in the Inducible Bacteriocin-producing Lactobacillus Plantarum Strains
【摘要】 通过共培养诱导产生细菌素,成功筛选到两株植物乳杆菌XJ25和PC520。基于两株菌基因组重测序,分析其植物乳杆菌素(plantaricin,pln)基因座间存在的差异。结果:菌株XJ25和PC520的pln基因座结构相似,均由6个操纵子(plnEFI、plnJKLR、plnGHSTUVW、plnABCD、plnMNOP、plnWXY)组成,其中plnABCD是3组分调节操纵子。菌株XJ25和PC520均存在一个新的未知功能的orf1;菌株PC520的部分基因(plnY,plnS)等缺失,菌株XJ25的plnF基因编码蛋白在14位(A-S)和42位(V-I)有两个氨基酸突变。
【Abstract】 Lactobacillus plantarum XJ25 and PC520 strains produced bactericin successfully through the induction of co-culture. Based on the sequencing draft genome, the plnloci of XJ25 and PC520 were studied and compared. The study showed that the plnloci of XJ25 and PC520 were similar and comprised six operons, i.e. plnEFI, plnJKLR,plnGHSTUV, plnABCD, plnMNOP and plnWXY. And a new orf1 was also found with unknown function. The regulatory operon of the two strains was plnABCD with three components. Moreover, plnY and plnS genes were missing in PC520 plnlocus while on the protein encoded by XJ25 plnF gene, mutations of two amino acids(A-S, V-I) were detected at sites 14 and 42, respectively.
