鸡胚发育早期肌肉中脂肪沉积规律的研究

【摘要】 为研究鸡胚胎发育早期肌肉中脂肪沉积的变化规律,试验选择玫瑰冠鸡和科宝鸡为研究对象,通过对6~13胚龄腿肌、胸肌组织压片和油红O染色,结合Image J软件统计分析肌肉中脂滴沉积量。结果表明:两种鸡腿肌均在7胚龄(E7)开始出现脂肪沉积,胸肌则在8胚龄(E8)开始出现脂肪沉积,玫瑰冠鸡腿肌在E8、E10、E11和E12胚龄的脂滴沉积量均极显著高于科宝鸡(P<0.01),E9期显著低于科宝鸡(P<0.05);玫瑰冠鸡胸肌在E10期和E11时的脂滴沉积量显著低于科宝鸡(P<0.05),在E12期极显著高于科宝鸡(P<0.05)。玫瑰冠鸡腿肌和胸肌E13期的脂滴沉积量均极显著低于科宝鸡(P<0.01),且两种鸡腿肌的脂滴含量达到最低,而胸肌则达到最高。综上所述,鸡胚胎发育早期腿肌中的脂肪沉积早于胸肌,两种鸡腿肌的脂肪沉积量随胚龄增长呈先升高再下降的趋势,且玫瑰冠鸡在E12期前大多高于科宝鸡,两种鸡胸肌的脂肪沉积量随胚龄增长呈升高-降低-升高的规律。

【Abstract】 In order to study the changes of lipid deposition in the early stage of chicken embryo development, the experiment selected Rose-crown chicken and Cobb chicken as the research object, the amount of lipid deposited in muscle was analyzed through leg and breast muscle tissue compression slices and oil red O staining at 6 to 13 days of embryonic age, combined with image J software. The results showed that the lipid began to appear in the leg muscles of both breeds at 7 days of embryonic age, and the breast muscles began to appear lipid at days of 8 embryonic age(E8). The lipid deposition in leg muscle of Rose-crown chicken at E8, E10, E11 and E12 were significantly higher than those of Cobb chicken(P<0.01), which was significantly lower than that of Cobb chicken at E9(P<0.05). The lipid deposition in breast muscle of Rose-crown chicken was significantly lower than that of Cobb chicken at E10 and E11(P<0.05), and significantly higher than that of Cobb chicken at E12(P<0.05). The lipid deposition in leg and breast muscle of Rose-crown chicken at E13 was significantly lower than that in Cobb chicken(P<0.01), the lipid droplets in legs of the two breeds reached the lowest, while there was the highest in the chest muscle. In summary, the fat deposition in leg muscle was earlier than in breast muscle at the early embryonic development of chickens. The fat deposition in leg muscles of two breeds increased firstly and then decreased with the growth of the embryonic age, and Rose-crown chicken were mostly higher than Cobb chicken before E12. The fat deposition of breast muscle of the two breeds increased firstly, then decreased, and then increased.
