鸡miRNA let-7b靶向调控GHR基因的双荧光素酶验证
Functional Verification of MiRNA Let-7b Targeted GHR Gene by Double Luciferase Reporter System
【摘要】 miRNA是一类大小约为22 nt的内源性非编码小RNA,通过与其靶基因mRNA相结合而影响其功能,进而调控动物的生长发育过程。miRNA靶基因预测软件发现,生长激素受体(GHR)基因是miRNA let-7b的候选靶基因之一。GHR基因作为生长激素基因的受体基因,在鸡的生长发育过程中具有极其重要的作用。试验采用双荧光素酶报告系统对其靶向关系进行验证。结果表明:GHR mRNA 3′UTR存在与let-7b相结合的靶标位点(UACCUCA);Let-7b过表达载体与GHR验证载体共转染组的荧光素酶相对活性只有0.159,明显下降,表明miRNA let-7b靶向负调控GHR 3′UTR,存在确切的靶标关系。研究结果可为进一步揭示let-7b介导GHR基因在鸡生长发育过程中的调控作用提供参考。
【Abstract】 MiRNA is a class of endogenous non-coding small RNA molecule with a size of about 22 nt that directly regulates the switching of certain genes to control the growth and development of animals. The target gene prediction software was found that the growth hormone receptor(GHR) gene was one of the candidate target genes for let-7b. As a receptor gene of growth hormone gene, GHR gene plays an extremely important role in the growth and development of chicken. This experiment used a dual luciferase reporter system to verify its targeting relationship. The results showed that the 3′UTR of GHR mRNA had a target site binding to let-7b(UACCUCA). The relative activity of luciferase in the co-transfection group of let-7b overexpression vector and GHR validation vector was only 0.159, which was significantly decreased. It was indicated that miRNA let-7b targets regulated GHR 3′UTR negatively, and there was an exact target relationship. This results could provide a reference for further revealing the regulation of let-7b-mediated GHR gene in growth and development of chicken.
