整合素αvβ3在人工受精奶牛受孕前后表达规律研究

【摘要】 本试验通过对10头荷斯坦奶牛在受孕前后的整合素αvβ3表达变化规律与奶牛早期妊娠关系的研究,筛选早期妊娠检测标志性因子,从而提高奶牛的繁殖效率。试验表明,选取的10头荷斯坦奶牛中有7头受孕3头未受孕,检测发现,10、13、16、19d,αv基因和β3基因转录及整合素αvβ3表达水平在受孕奶牛血液中的表达量均高于未孕奶牛,特别是输精后第16d受孕奶牛的表达量显著高于未孕奶牛。因此,αvβ3检测可将早孕检测的时间窗口提前到人工输精后第16d。

【Abstract】 This experiment was conducted to study the relationship between the expression of integrin αvβ3 in 10 Holstein cows before and after conception and the early pregnancy of dairy cows, and to screen the early pregnancy detection marker factors, so as to improve the reproductive efficiency of dairy cows. Ten cows were selected with similar birth weight and healthy Holstein cows in a dairy farm in Beijing in 2016, and this pregnancy was a second child, with estrus and insemination on the same day. Blood samples were collected on day 10, 13, 16 and 19 after insemination, and the changes and regularities of integrin αvβ3 and its mRNA levels were analyzed by realtime PCR and Western blot. Seven of the 10 Holstein cows were conceived and others were not conceived. It was found that the expression levels of αv gene and β3 gene and integrin αvβ3 were in the blood of pregnant cows at day 10, 13, 16 and 19. The expression was higher than that of the non-pregnant cows, especially on the day 16 after insemination, which was significantly higher than that of the non-pregnant cows. The time window of early pregnancy detection can be advanced to the day 16 after artificial insemination by αvβ3 detection.
