水稻条纹花叶病田间调查及分子检测

【摘要】 水稻条纹花叶病(Rice stripe mosaic disease)是近年在华南地区新发现的水稻病毒病。该病由电光叶蝉(Recilia dorsalis)以持久增殖型方式传播。2017年至2018年,连续两年的调查结果表明,该病害广泛分布于华南稻区,以广东西南和广西东南部发生较重。室内人工接种和田间病株观察揭示,该病的症状主要表现为病株轻度矮化,叶片呈淡黄色条纹或花叶,重病株叶片基部扭转畸形,抽穗不完全,籽粒不饱满。根据病毒复制酶(L)基因设计一对特异引物(RSMV-L-F:5′-CTCCAACTATCATCCGCTATGC-3′;RSMV-L-R:5′-CCATCCGAGATAAGGTCACTGT-3′),经RT-PCR可对水稻病株及叶蝉体内的病原病毒进行快速、准确的检测。

【Abstract】 Rice stripe mosaic disease discovered recently was a new viral disease affecting rice in Southern China. This disease was transmitted by a leafhopper Recilia dorsalis with persistent propagative manner. In this study, field investigations during the year 2017 to 2018 revealed that it was widely distributed in Southern China, and occurred seriously in the southwest of Guangdong and the southeast of Guangxi. Artificial inoculation and field observation showed that symptoms of this disease included plant mild dwarf, leaves mosaic, yellow stripe and distortion and incomplete heading. A pair of specific primers based on the viral replicase gene(RSMV-L-F:5′-CTCCAACTATCATCCGCTATGC-3′;RSMV-L-R:5′-CCATCCGAGATAAGGTCACTGT-3′) was designed and an RT-PCR method was established successfully for detection of the virus.
