5-氨基酮戊酸光动力疗法对兔耳痤疮模型皮损及组织中IL-17表达水平的影响
Effects of 5-aminolevulinic Acid Photodynamic Therapy on the General Appearance of Rabbit Ear Acne Model and the Expression of IL-17
【摘要】 目的探讨5-氨基酮戊酸光动力疗法(5-aminolevulinicacid photodynamic therapy,ALA-PDT)对兔耳痤疮模型皮损治疗效果和ALA-PDT治疗前后皮损组织中IL-17表达水平及初步机制研究。方法将痤疮丙酸杆菌(Propionibacterium acnes,P.acnes)皮内注射于兔耳部皮肤,建立痤疮动物模型。建模后动物分为:ALA-PDT组及无干预组。通过观察皮损大体表现及其组织病理改变评价痤疮炎症变化趋势;采用Real-time PCR技术检测ALA-PDT治疗前以及治疗后不同时间点(4 h、24 h、4 d、7 d)IL-17及其上游转录因子RORα和RORγt的表达水平。结果兔耳皮肤注射P.acnes菌悬液可诱导痤疮样皮损。皮损大体表现,ALA-PDT组在治疗后4~24 h皮损区炎症逐渐加重;治疗后4 d组织淋巴细胞浸润较4 h、24 h减轻;治疗后7 d皮损好转明显且优于无干预组。Real-time PCR研究结果发现ALA-PDT组IL-17、RORα、RORγt表达水平在治疗后4 h逐渐升高,24 h后进一步升高;治疗后4 d表达水平较前降低;治疗后7 d表达水平显著降低。结论 ALA-PDT早期通过上调IL-17放大炎症反应,随后下调IL-17,最终有效治疗痤疮。ALA-PDT的这一调控作用可能通过两个关键转录因子RORα和RORγt实现。
【Abstract】 Objective To explore the effect of 5-aminolevulinicacid photodynamic therapy(ALA-PDT) on the rabbit ear acne(REA) model treatment and the expression of IL-17 and its preliminary regulation mechanism.Methods P.acnes was injected intradermally into the skin of rabbit ears to establish a rabbilt ear ance model.Model animals were divided into ALA-PDT group and no treatment group.The variation trend of tissue inflammatory response was evaluated by observing the general appearance and conducting HE stain.Real-time PCR was used to test the mRNA expression level of IL-17 and its upstream transcription factor RORα and RORγt before and at different time points(4 h,24 h,4 d,7 d) after ALA-PDT treatment.Results REA model can be induced by injecting P.acnes suspension into rabbit ear skin.By observing the general appearance,the inflammation performance of skin lesion in the ALA-PDT group was gradually aggravated from 4 h to 24 h after the treatment.It was observed an alleviative inflammation response in the fourth day after the treatment in contrast to 4 h and 24 h.After seven days of the treatment,the skin lesions in ALA-PAT group disappeared significantly and was conspicuously better than the no treatment control group.Real-time PCR showed that the expression levels of IL-17,RORα,RORγt mRNA in ALA-PDT treatment group increased gradually after 4 h of treatment and a further increase after 24 h.They decreased after four days of treatment than before.After seven days,the expression levels of the three factors were significantly lower than that before treatment.Conclusion ALA-PDT amplifies the inflammatory response by first up-regulating IL-17,then down-regulating IL-17,ultimately curing acne.This effect of ALA-PDT on IL-17 may be achieved by regulating two key transcription factors,RORα and RORγt.
