【摘要】 目的:建立白木香叶的高效液相分析方法并对其进行方法学考察,为白木香叶指纹图谱研究提供依据。方法:分别对检测波长、流动相系统、流动相洗脱梯度、供试品溶液进样体积、流速及色谱柱柱温进行考察,确定最终色谱条件。从专属性、仪器精密度、重复性及方法耐用性等方面考察所建立的分析方法。结果:白木香叶的最终色谱条件如下,色谱柱为Shiseido CAP Cell MGⅢC₁₈柱（250 mm×4.6 mm,5μm）,流速为1.0 ml/min,检测波长为280 nm,柱温为30℃,进样体积为10μl,流动相为乙腈-0.1%磷酸水溶液,梯度洗脱。方法学考察结果表明,本方法专属性好、仪器精密度符合要求、重复性符合要求且耐用性稳定。结论:建立的白木香叶高效液相色谱分析方法能满足方法学考察要求,且操作简便,可应用于白木香叶的高效液相指纹图谱分析。更多还原

【Abstract】 OBJECTIVE: To establish the HPLC method for aquilaria sinensis leaves and its methodological investigation, so as to provide basis for the study of fingerprints of aquilaria sinensis leaves. METHODS: The detection wavelength, mobile phase system, mobile phase elution gradient, sample solution injection volume, flow rate and column temperature were investigated to determine the final chromatographic conditions. The analysis method was investigated from the aspects of specificity, instrument precision, repeatability and method durability. RESULTS: The chromatographic conditions of HPLC analysis of aquilaria sinensis leaves were finally determined, the column was Shiseido CAP Cell MG Ⅲ C₁₈ column（250 mm×4.6 mm, 5 μm） with flow rate of 1.0 ml/min, detection wavelength of 280 nm, column temperature of 30 ℃, injection volume of 10 μl, and mobile phase of acetonitrile-0.1% aqueous phosphoric acid and gradient elution. The results of the methodological investigation showed that the method had good specificity, the precision of the instrument met the requirements, the repeatability met the requirements and the durability was stable. CONCLUSIONS: The established HPLC method for aquilaria sinensis leaves can meet the requirements of methodological investigation, and is easy to operate, which can be applied to HPLC analysis of aquilaria sinensis leaves.更多还原
