高效液相色谱法测定不同产地蒲黄药材中3种黄酮苷元含量
Content Determination of Three Flavonoid Aglycones in Typhae Pollen from Different Habitats by HPLC
【摘要】 目的建立测定不同产地蒲黄药材中槲皮素、山柰素和异鼠李素3种黄酮苷元含量的高效液相色谱(HPLC)法。方法色谱柱采用Hypersil BDS-C18柱(250 mm×4. 6 mm,5μm),流动相为甲醇-0. 4%磷酸溶液(50∶50,V/V),流速为1. 0 mL/min,柱温为30℃,检测波长为360 nm,进样量为10μL。结果蒲黄药材中槲皮素、山柰素、异鼠李素进样量线性范围分别为40. 12~1 002. 89 ng(r=1. 000 0),21. 70~542. 42 ng(r=0. 999 9),59. 90~1 497. 38 ng(r=1. 000 0),平均回收率分别为96. 45%,94. 08%,99. 11%,RSD分别为2. 49%,1. 25%,1. 32%(n=6)。不同产地蒲黄中3种黄酮苷元的含量差异较大,其中广西桂林产最高,槲皮素为0. 372 1 mg/g,山柰素为0. 417 3 mg/g,异鼠李素为1. 702 8 mg/g;内蒙古包头和宁夏固原产相对较低。结论该方法快速、准确,可用于蒲黄药材中3种黄酮苷元的含量测定。
【Abstract】 Objective To establish an HPLC method for the content determination of three flavonoid aglycones( quercetin,kaempferin and isorhamnetin) in Typhae Pollen from different habitats by HPLC. Methods The chromatographic column was Hypersil BDS-C18 column( 250 mm × 4. 6 mm,5 μm),the mobile phase was methanol-0. 4% phosphoric acid solution( 50 ∶ 50,V/V),the flow rate was1. 0 mL/min,the column temperature was 30 ℃,the detection wavelength was 360 nm,and the sample size was 10 μL. Results The linear range was 40. 12-1 002. 89 ng( r = 1. 000 0) for quercetin,21. 70-542. 42 ng( r = 0. 999 9) for kaempferin and 59. 90-1 497. 38 ng( r = 1. 000 0) for isorhamnetin. The average recoveries of quercetin,kaempferin and isorhamnetin were 96. 45%,94. 08%and 99. 11%,and their RSDs were 2. 49%,1. 25% and 1. 32%( n = 6). The contents of three flavonoid aglycones in Typhae Pollen from different habitats were quite different. Among them,the contents of three flavonoid aglycones in Typhae Pollen from Guilin,Guangxi were the highest: the content of quercetin was 0. 372 1 mg/g,the content of kaempferin was 0. 417 3 mg/g,and the content of isorhamnetin was 1. 702 8 mg/g. The contents of flavonoid aglycones in Typhae Pollen from Baotou,Inner Mongolia and Guyuan,Ningxia were relatively low. Conclusion The method is rapid and accurate,which can be used for the content determination of three flavonoid aglycones in Typhae Pollen.
