SPOCK1在大肠癌组织中表达的研究

【摘要】 目的对SPOCK1 mRNA及其蛋白在大肠癌组织、癌旁组织中的表达和SPOCK1蛋白质的表达与临床病理特征之间的关系进行探讨。方法收集哈尔滨医科大学附属第四医院普外科2014年7月1日~2015年7月1日手术切除的19例新鲜大肠癌组织及癌旁组织标本(癌旁组织标本为正常的大肠黏膜组织,距离癌组织≥5cm),每例标本取2块,1块用于实时荧光定量PCR(real-time-PCR)检测大肠癌组织中SPOCK1 mRNA的表达情况,另外1块制成石蜡标本,连同收集的笔者医院病理科2007年7月1日~2012年7月1日大肠癌及癌旁石蜡标本35例,共54例标本采用免疫组织化学方法(immuno hisochemistry,IHC)检测SPOCK1蛋白的表达,将结果与相应的临床病理学资料进行统计学数据分析。结果实时荧光定量PCR结果显示,在大肠癌组织标本中的SPOCK1 mRNA比癌旁组织中的表达高,差异有统计学意义(P<0.05);IHC结果显示,在大肠癌组织标本中的SPOCK1蛋白比癌旁组织中的表达高,差异有统计学意义(P<0.05)。SPOCK1蛋白与临床病理学资料分析结果显示,SPOCK1蛋白的表达和大肠癌组织分级呈正相关,差异有统计学意义(P<0.05)。结论 SPOCK1在大肠癌组织中呈现高表达,其表达量同大肠癌组织学分级呈正相关。

【Abstract】 Objective To investigate the expression of SPOCK1 mRNA and its protein in colorectal cancer tissues, and adjacent tissues, and at the same time to analyze the relationship between SPOCK1 protein and clinical pathology features. Methods We collected 19 cases of colorectal cancer specimens and adjacent tissues(from the cancer tissue ≥5 cm) from department of general surgery of the Fourth Hospital of Harbin Medical University. Each sample was divided into 2, in which 1 was used to detect the expression of SPOCK1 mRNA in colorectal cancer tissues by real-time fluorescence quantitative PCR. The other part was prepared into paraffin tissue and 35 cases of paraffin tissue collected from pathology department of the Fourth Hospital of Harbin Medical University were used to detect the expression of SPOCK1 protein by immunohistochemistry(IHC). And we carried out statistical analysis with clinical data. Results Real-time fluorescence quantitative PCR showed that SPOCK1 mRNA was significantly higher in colorectal cancer than in adjacent tissues(P<0.05), and the difference was statistically significant. IHC results showed that the expression of SPOCK1 protein was higher in colorectal cancer than in normal tissue(P<0.05), and the difference was statistically significant(P<0.05). The results of clinicopathological data showed that the expression of SPOCK1 protein was positively correlated with the grade of colorectal cancer(P<0.05). ConclusionSPOCK1 was highly expressed in colorectal cancer tissues, and its expression was positively correlated with histological grade of colorectal cancer.
