脂肪间充质干细胞治疗慢性阻塞性肺疾病的实验研究
Application of Adipose Derived Mesenchymal Stem Cells in the Treatment of Chronic Obstructive Pulmonary Disease
【摘要】 目的探究脂肪间充质干细胞(ADMSCs)在慢性阻塞性肺疾病(COPD)治疗中的价值。方法将60只SPF级雄性SD大鼠随机分为对照组、治疗组及模型组,每组20只,治疗组和模型组采用熏香烟加气管注入内毒素进行COPD模型的制备,对照组不造模。造模后治疗组大鼠尾静脉注射0.5 ml溴氮胞苷(BrdU)标记的ADMSCs悬液,对照组及模型组大鼠经尾静脉注射0.5ml生理盐水。移植后14d,测定各组肺功能、炎症因子水平及肺组织病理变化。结果与对照组比较,模型组和治疗组肺功能指标均显著下降(P<0.05),而治疗组肺功能优于模型组(P<0.05);模型组和治疗组肺泡灌洗液中白细胞总数、巨噬细胞数、中性粒细胞数、白细胞介素-8、肿瘤坏死因子及C反应蛋白水平均显著高于对照组(均P<0.05),而治疗组上述各指标均显著低于模型组(P<0.05);模型组、治疗组肺组织切片均可见COPD病理性变化,治疗组较模型组肺气肿病理性变化轻(P<0.05)。结论 ADMSCs移植治疗可显著改善COPD大鼠肺组织的病理性改变,减轻炎症反应,改善肺功能。
【Abstract】 Objective To explore the application value of adipose derived mesenchymal stem cells(ADMSCs)in chronic obstructive pulmonary disease(COPD).Methods 60 SPF male rats were randomly divided into control group,observation group and model group with 20 rats in each group.COPD rat models were established in observation group and model group by passive cigarette smoking and intratracheal instillation of endotoxin, while non model was established in control group. After modeling,rats in observation group were injected with 0. 5 ml of BrdU labeled ADMSCs suspension through caudal vein,while rats in control group and model group were injected with 0.5 ml of normal saline through caudal vein.Lung function,levels of inflammatory factors and pathological changes of lung tissue in rats were determined at 14 d after transplantation.Results Compared with the control group,the indexes of lung function decreased significantly in model group and observation group(P<0.05),and lung function in observation group was better than that in model group(P<0.05).The number of total white blood cells,number of macrophages and neutrophils,levels of interleukin 8,tumor necrosis factor and C reactive protein in bronchoalveolar lavage fluid of the model group and observation group were significantly higher than those in control group(P<0.05).The number of immune cells and levels of inflammatory factors in observation group were significantly lower than those in model group(P<0.05).Pathological changes of COPD in the lung tissue sections of model group and observation group were observed.Emphysema symptoms in observation group were milder than those in model group(P<0.05).Conclusion ADMSCs transplantation can sig nificantly improve pathological changes of lung tissues in COPD rats,relieve inflammatory reaction and improve lung function.
