糖代谢与抗肿瘤治疗
Glucose metabolism and anti-tumor therapy
【摘要】 能量代谢重编程是肿瘤的十大特征之一,其中葡萄糖代谢异常是肿瘤代谢最突出的特征。在氧气充足的情况下,肿瘤细胞依然倾向于进行糖酵解,将葡萄糖代谢为乳酸。肿瘤细胞有氧糖酵解能力是正常细胞的20~30倍,为肿瘤代谢提供大量能量和中间产物。因此,靶向糖酵解等异常环节的代谢酶是抗肿瘤治疗的重点,目前相关研究也取得了一定的进展。本文综述了肿瘤中的糖代谢异常调控及靶向相关代谢反应的抑制剂研究进展。
【Abstract】 Reprogramming of energy metabolism is one of the hallmarks of cancer, among which glucose metabolism dysfunction is the most prominent feature. Under the condition of suf?cient oxygen, cancer cells still undergo high rates of aerobic glycolysis,metabolizing glucose to lactate. The aerobic glycolysis capacity of tumor cells is 20~30 times that of normal cells, providing large amount of energy and intermediate products. Therefore, targeting the dysregulation process of glycolysis is important for antitumor therapy, and the related drug discovery study has made signi?cant progress. Here we reviewed the reprogramming of glucose metabolism in tumor and the target therapy studies.
