UPLC法同时测定降香中7个黄酮类成分含量及主成分分析
Simultaneous determination of seven flavonoids in Dalbergiae Odoriferae Lignum by UPLC and principal component analysis
【摘要】 目的:建立同时测定市售降香中甘草素、异甘草素、紫铆花素、柚皮素、漆黄素、芒柄花素和鹰嘴豆芽素A含量的UPLC法,并应用主成分分析法评价其质量。方法:使用甲醇超声提取降香药材中上述7个黄酮类成分,并应用超高效液相色谱法测定其含量。色谱条件:采用ACQUITY BEH C18色谱柱(1.7μm,3.0 mm×100 mm),柱温35℃,以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速0.3 mL·min-1</sup>,检测波长270 nm和370 nm;使用统计学软件SPSS 22.0,对样品中7个黄酮类成分含量测定结果进行主成分分析。结果:甘草素、异甘草素、紫铆花素、柚皮素、漆黄素、芒柄花素和鹰嘴豆芽素A的线性范围分别为0.014 0~0.140 0μg(r=0.999 9)、0.008 3~0.083 0μg(r=0.999 9)、0.008 1~0.081 0μg(r=0.999 9)、0.005 8~0.058 0μg(r=0.999 9)、0.008 2~0.082 0μg(r=0.999 9)、0.025 5~0.255 0μg(r=0.999 9)、0.015 2~0.152 0μg(r=0.999 9);平均回收率(n=9)分别为98.7%、98.1%、98.5%、99.6%、98.9%、99.2%、100.9%,RSD分别为1.7%、2.1%、2.7%、2.6%、2.4%、1.4%、2.3%;降香样品中上述7个黄酮类成分含量范围分别为0.19~6.41、0~0.45、0~1.03、 0~12.61、0.12~2.06、0.59~5.96、0~4.86 mg·g-1。由主成分分析可知,选择3个因子(F1、F2、F3)对不同来源降香药材进行综合评价,综合评价函数为F=43.840F1+26.436F2+13.250F3,其中,海南产降香药材,相对于其他地方产的药材质量较好。结论:该方法操作简便、快捷,结合主成分分析,可以对不同来源降香质量进行客观、有效的评价。
【Abstract】 Objective:To establish an UPLC method for simultaneous determination of liquiritigenin,isoliquiritigenin,buturisin,naringenin,fisetin,formononetin,and biochanin A in Dalbergiae Odoriferae Lignum,and to evaluate the quality of the drug with principal component analysis. Methods:The above seven flavonoids were extracted by ultrasonic machine with methanol and the contents were determined by UPLC. The separation was performed on ACQUITY BEH C18(1.7 μm,3.0 mm×100 mm)and column temperature was 35 ℃. The mobile phase consisted of acetonitrile and 0.1% phosphoric acid with gradient elution at a flow rate of 0.3 mL·min-1</sup>. The detection wavelengths were set at 270 nm and 370 nm. The results were analyzed by SPSS 22.0. Results:The linear ranges of liquiritigenin,isoliquiritigenin,buturisin,naringenin,fisetin,formononetin,and biochanin A were 0.014 0-0.140 0 μg(r=0.999 9),0.008 3-0.083 0 μg(r=0.999 9),0.008 1-0.081 0 μg(r=0.999 9),0.005 8-0.058 0 μg(r=0.999 9),0.008 2-0.082 0 μg(r=0.999 9),0.025 5-0.255 0 μg(r=0.999 9)and 0.015 2-0.152 0 μg(r=0.999 9),respectively. The average recovery rates(n=9)were 98.7%,98.1%,98.5%,99.6%,98.9%,99.2%,and 100.9%,with RSD of 1.7%,2.1%,2.7%,2.6%,2.4%,1.4% and 2.3%,respectively. In Dalbergiae Odoriferae Lignum from different sources,the content ranges of liquoricein,isoliquiritigenin,buturisin,naringenin,fisetin,formononetin,and biochanin A were 0.19-6.41,0-0.45,0-1.0,0-12.61,0.12-2.05,0.59-5.96 and 0-4.86 mg·g-1,respectively. The principal components analysis showed that three factors(F1,F2,F3)could be used for comprehensive evaluation with the function of F=43.840 F1+26.436 F2+13.250 F3. The quality of samples from Hainan was better than others. Conclusion:The UPLC method is simple and quick to operate. Combined with principal component analysis,it can evaluate the quality of Dalbergiae Odoriferae Lignum in an effective and objective way.
