右美托咪定通过降低IL-17和RORγt的表达降低脓毒症小鼠的肺损伤

【摘要】 目的观察盲肠结扎法对小鼠血液和肺组织中白细胞介素(IL-17)和维甲酸相关孤核受体γt(RORγt)表达的影响和右美托咪定的干预作用。方法雄性C57BL/6小鼠32只,体重20~25g,随机分为四组:S组(n=8)为假手术组;Dex组(n=8)为假手术加右美托咪定40μg/kg腹腔注射;Sep组(n=8)为盲肠结扎穿孔组;Sep+Dex组(n=8)为盲肠结扎穿孔加右美托咪定40μg/kg腹腔注射。24h后用眼球放血处死小鼠,并收集小鼠的血液和肺组织。采用苏木精-伊红法(HE)染色后,在光镜下观察肺组织损伤评分;检测小鼠肺湿重/干重比(W/D);ELISA法检测血浆和肺组织中IL-17的浓度;Western blot法检测肺组织RORγt蛋白的含量。结果 S组和Dex组未见明显损伤,Sep组肺出血、充血、炎性细胞浸润明显,Sep+Dex组肺损伤程度明显减轻(P<0.01)。与S组比较,Sep组小鼠外周血和肺组织W/D明显升高,IL-17浓度明显升高,肺组织中RORγt蛋白含量明显升高(P<0.05)。与Sep组比较,Sep+Dex组上述指标都明显降低(P<0.05)。结论盲肠结扎模型可导致小鼠外周血和肺组织IL-17及肺组织RORγt的表达水平明显升高,而右美托咪定可通过降低IL-17和RORγt的表达减轻盲肠结扎致脓毒症小鼠肺损伤程度。

【Abstract】 Objective To observe the effect of cecal ligation on the expressions of interleukin(IL)-17 and retinoic-acid-related orphan receptorγt(RORγt)in blood and lung tissues of mice and the intervention effect of dexmedetomidine.Methods Thirty-two male C57 BL/6 mice(weighed 20~25 g)were randomly divided into four groups:group S(n =8)was sham operation group;group Dex(n =8)was sham operation plus intraperitoneal injection of dexmedetomidine 40μg/kg;group Sep(n =8)was cecal ligation perforation group;group Sep+ Dex(n =8)was cecal ligation perforation plus intraperitoneal injection of dexmedetomidine 40μg/kg.After 24 hours,the mice were killed by eyeball bleeding,and the mouse blood and lung tissues were collected.After hematoxylin-eosin(HE)staining,the lung injury score was observed by light microscope;the ratio of wet weight to dry weight(W/D)of the mouse lung was detected;the concentration of IL-17 in plasma and lung tissues was detected by ELISA;and the content of RORγt protein in lung tissues was detected by Western blot.Results There was no obvious injury in group S and group Dex,and the pulmonary hemorrhage,hyperemia and inflammatory cell infiltration were obvious in group Sep,and the degree of lung injury in group Sep+Dex was significantly reduced(P<0.01).Compared with group S,W/D in peripheral blood and lung tissues of mice in group Sep was significantly increased,IL-17 concentration was significantly increased,and the RORγt protein content in lung tissue was significantly increased(P<0.05).Compared with group Sep,the above indexes in group Sep+Dex were significantly lower(P<0.05).Conclusion The expressions of IL-17 and RORγt in mouse peripheral blood and lung tissues can be significantly increased in cecal ligation model,while dexmedetomidine can reduce the degree of lung injury in sepsis mice by reducing the expressions of IL-17 and RORγt.
