基于数字信息学方法筛选软骨肉瘤细胞中的抑癌MicroRNA及其对软骨肉瘤细胞增殖和侵袭能力的影响

【摘要】 目的:筛选软骨肉瘤细胞中的抑癌MicroRNA,研究其对软骨肉瘤细胞迁移、侵袭、增殖和生长能力的影响,为进一步探究其作用机理夯实基础。方法:通过筛选30种文献报道最多的miRNA在人软骨肉瘤组织和细胞中的表达情况,并通过数字信息学方法绘制热点图,获得人软骨肉瘤中显著差异表达的目的miRNA——miR-140-5p;RT-PCR确认miR-140-5p在62例人软骨肉瘤组织及3例人软骨肉瘤细胞系JJ012的表达情况;上调miR-140-5p对JJ012细胞迁移、侵袭、增殖能力的影响;通过裸鼠皮下成瘤实验明确miR-140-5p对JJ012体内成瘤能力的作用。结果:miR-140-5p在软骨肉瘤中的低表达有显著差异性;调高miR-140-5p可明确抑制软骨肉瘤细胞的侵袭、迁移和增殖能力;在裸鼠皮下成瘤实验中,miR-140-5p在体内显著抑制软骨肉瘤的生长。结论:miR-140-5p在人软骨肉瘤组织及JJ012细胞中表达降低;上调miR-140-5p能显著抑制JJ012细胞迁移、侵袭、增殖和生长能力。
【Abstract】 Objective: To screen the anti-cancer MicroRNA in chondrosarcoma cells, study its effect on the migration, invasion, proliferation and growth ability of chondrosarcoma cells, and lay a solid foundation for further exploration of its mechanism of action. Methods: By screening the expression of miRNA in human chondrosarcoma tissues and cells reported most in 30 kinds of literatures and drawing the hotspot map with the method of digital informatics, miR-140-5 p, the target miRNA with significantly different expression in human chondrosarcoma is obtained; the expression of miR-140-5 p in 62 cases of human chondrosarcoma tissues and 3 cases of human chondrosarcoma cell line JJ012 is confirmed by RT-PCR; the effect of miR-140-5 p on migration, invasion and proliferation of JJ012 cells is up-regulated; and the effect of miR-140-5 p on tumorigenesis of JJ012 in vivo is determined by subcutaneous tumorigenesis experiment of nude mice. Results: There are significant differences in the low expression of miR-140-5 p in chondrosarcoma; up-regulation of miR-140-5 p can significantly reduce the invasion, migration and proliferation ability of chondrosarcoma cells; and in the subcutaneous tumorigenesis experiment of nude mice, miR-140-5 p significantly inhibited the growth of chondrosarcoma in vivo. Conclusion: The expression of miR-140-5 p decreases in human chondrosarcoma tissues and JJ012 cells. The up-regulation of miR-140-5 p can significantly reduce the migration, invasion, proliferation and growth ability of JJ012 cells.
