小鼠耳廓软骨细胞微囊化大鼠胰岛细胞制备软骨细胞薄膜人工生物胰对糖尿病小鼠血糖水平的影响
Effects of chondrocyte membrane artificial biological pancreas prepared by microencapsulating rat pancreatic islet cells with mouse auricular chondrocyte on blood glucose level in diabetic mice
【摘要】 [目的]观察小鼠耳廓软骨细胞微囊化大鼠胰岛细胞制备软骨细胞薄膜人工生物胰对糖尿病小鼠血糖水平的影响.[方法]取8~9周龄雄性C57BL/6小鼠,在无菌条件下切取两耳并分离耳廓软骨细胞,取第2代软骨细胞备用.实验分为两组:1)取耳廓软骨细胞与500~600个胰岛细胞共同加入6 cm Hydrocell Plate中进行振荡培养5 d,制备软骨细胞薄膜人工生物胰;2)在384孔Hanging Drop Plate内放入1个胰岛细胞和5 000个软骨细胞静培养3 d,制备软骨细胞薄膜人工生物胰.将两组人工生物胰分别移植入用链脲佐菌素制作的糖尿病模型小鼠的肾被膜下,观察移植后糖尿病模型小鼠血糖水平的变化.[结果]利用两种方法制备的软骨细胞薄膜人工生物胰均可稳定地将糖尿病模型小鼠血糖水平控制在正常范围内,其中利用Hydrocell Plate培养方法制备的软骨细胞薄膜人工生物胰回收量较利用Hanging Drop Plate培养方法制备的更多,实验人员工作量更少,且利用Hydrocell plate培养方法制备的软骨细胞薄膜人工生物胰降低血糖作用较利用Hanging Drop Plate培养方法制得的更为明显.[结论]小鼠耳廓软骨细胞微囊化大鼠胰岛细胞制备的软骨细胞薄膜人工生物胰可稳定地将糖尿病模型小鼠血糖水平控制在正常范围内.
【Abstract】 OBJECTIVE To observe the effects of chondrocyte membrane artificial biological pancreas prepared by microencapsulating rat islet cells with mouse auricular chondrocyte on blood glucose in diabetic mice. METHODS Male C57 BL/6 mice aged 8-9 weeks were selected, and their two ears were excised and auricular chondrocytes were isolated under sterile condition, and the second generation chondrocytes were taken for reserve. The experiment was divided into two groups:1) Auricular chondrocyte and 500-600 islet cells were cultured in 6 cm Hydrocell Plate for 5 days to prepare chondrocyte membrane artificial biological pancreas;2) Chondrocyte membrane artificial biological pancreas was prepared by adding 1 islet cell and 5 000 auricular chondrocyte into Hanging Drop Plate with 384 holes and culturing for 3 days. Two groups of artificial biological pancreas were transplanted under the renal capsule of diabetic mice prepared with streptozotocin to observe the changes of blood glucose level after transplantation. RESULTS The chondrocyte membrane artificial biological pancreas prepared by the two culture methods could stably control the blood glucose level of diabetic mice in the normal range. The recycling amount of chondrocyte membrane artificial biological pancreas prepared by Hydrocell Plate culture method was more than that by Hanging Drop Plate culture method, and the workload of the experiment was less. Moreover, the hypoglycemic effect of chondrocyte membrane artificial biological pancreas prepared by Hydrocell Plate was more obvious than that prepared by Hanging Drop Plate. CONCLUSION Chondrocyte membrane artificial biological pancreas prepared by microencapsulating rat islet cells with mouse auricular chondrocyte could stably control the blood glucose level of diabetic mice within the normal range.
