CNP及其受体NPR-B在缺血再灌注损伤模型大鼠肾组织中的表达
Expressions of CNP and its receptor NPR-B in renal tissue of rats with ischemia reperfusion
【摘要】 [目的]观察CNP及其受体NPR-B在缺血再灌注损伤(IR)模型大鼠肾组织中的表达情况.[方法]取雄性Wistar大鼠随机分为假手术组(Sham组)、缺血再灌注0 h组(IR0 h)、1 h组(IR1 h)、2 h组(IR2 h)及4 h组(IR4 h).摘除各组大鼠左侧肾脏,Sham组分离右肾动静脉但不夹闭,IR各组分离右肾动静脉夹闭45 min后取下血管夹恢复肾脏血液供应,建立肾脏IR模型;各组大鼠到达各自时间段立即采集血液测定血清肌酐(Cr)及尿素氮(BUN)值,取各组大鼠肾组织观察NPR-B的分布及CNP和NPR-B mRNA表达情况.[结果]与Sham组比较,IR各组Cr,BUN水平明显升高(P<0.05),且随着再灌注时间的延长升高更为明显;NPR-B分布明显增多(P<0.05),CNP mRNA,NPR-B mRNA表达均显著升高(P<0.05),随着缺血再灌注时间的延长CNP mRNA表达亦随之增加,缺血再灌注4 h时表达水平最高.[结论]肾组织中CNP及NPR-B表达上调的机制认为可能是IR的代偿性保护作用.
【Abstract】 OBJECTIVE To observe the expressions of CNP and its receptor NPR-B in renal tissue of rats with ischemia reperfusion. METHODS Male Wistar rats were randomly divided into the Sham group,ischemia-reperfusion 0 h(IR0 h), ischemia-reperfusion 1 h(IR1 h), ischemia-reperfusion 2 h(IR2 h) and ischemia-reperfusion 4 h(IR4 h) group. All rats were left unilaterally nephrectomized. The right renal artery and vein were separated in the Sham group without clamping. In the IR groups, the right renal artery and vein were separated and clamped for 45 min, then the vascular clip was removed to restore the renal blood supply, and the renal IR model was established. The serum creatinine(Cr) and blood urea nitrogen(BUN) in the rats of each group was measured immediately at their respective time periods. The renal tissue of each group was taken to observe the distribution of NPR-B and the expressions of CNP and NPR-B mRNA. RESULTS Compared with the Sham group, the levels of Cr and BUN were significantly increased in the IR groups(P<0.05), and the increase was more obvious with the extension of reperfusion time, the distribution of NPR-B was significantly increased(P<0.05), and the expressions of CNP and NPR-B mRNA were significantly increased(P<0.05). The expression of CNP mRNA increased with reperfusion time, and the expression was highest at 4 h after ischemia reperfusion. CONCLUSION The mechanism of upregulation of CNP and NPR-B in renal tissue might be the compensatory protective effect of IR.
