TNF-α与β1,4-GalT-Ⅰ在膝骨关节炎滑膜炎症过程中的关系研究

【摘要】 目的探讨TNF-α与β1, 4-GalT-Ⅰ在膝骨关节炎(KOA)滑膜炎症过程中的关系。方法选取我院2016年3月至2018年2月诊断为KOA的15例患者作为观察组,选取同期15例健康检查者作为对照组,检测两组研究对象的滑膜组织TNF-α与β1, 4-GalT-Ⅰ的表达水平,并培养FLS,以酶联免疫吸附试验检测TNF-α的表达,以Real-time PCR检测β1, 4-GalT-Ⅰ的表达。结果观察组的TNF-α与β1, 4-GalT-Ⅰ相对表达量均明显高于对照组(P <0.05)。不同剂量TNF-α组的FLS,其β1, 4-GalT-Ⅰ相对表达量与对照组比较有统计学差异(P <0.05)。结论在KOA滑膜炎症过程中, TNF-α与β1, 4-GalT-Ⅰ参与了滑膜炎症的发生发展, TNF-α可能通过调控β1, 4-GalT-Ⅰ的表达参与KOA滑膜炎症。

【Abstract】 Objective To explore the relationship between TNF-α and β1, 4-GalT-Ⅰ in the process of synovial inflammation of knee osteoarthritis(KOA). Methods 15 cases of patients diagnosed as KOA in our hospital from March 2016 to February 2018 were selected as the observation group, and 15 people with health examination during the same period were selected as the control group. The expression levels of TNF-α and β1, 4-GalT-Ⅰ in synovial tissue of both groups were detected, and FLS were cultured to detect the expressions of TNF-α(ELISA) and β1, 4-GalT-Ⅰ(Real-time PCR). Results The relative expression levels of TNF-α and β1, 4-GalT-Ⅰ of the observation group were significantly higher than those of the control group(P <0.05). The relative expression level of β1, 4-GalT-Ⅰ in FLS with different doses of TNF-α had statistical difference with that of the control group(P <0.05). Conclusions In the process of KOA synovial inflammation, TNF-α and β1, 4-GalT-Ⅰ may be involved in the occurrence and development of synovial inflammation. TNF-α may be involved in the KOA synovial inflammation by regulating the expression of β1, 4-GalT-Ⅰ.
