美沙拉嗪联合酪酸梭菌对溃疡性结肠炎大鼠治疗作用及机制研究
Effects of mesalasize combined with clostridium butyricum in the treating experimental ulcerative colitis in rats and their potential mechanisms
【摘要】 目的研究美沙拉嗪联合酪酸梭菌对溃疡性结肠炎模型大鼠的治疗作用,并探讨其可能的作用机制。方法采用2,4,6-三硝基苯磺酸(TNBS)复合50%乙醇溶液对大鼠通过免疫化学法造模,将50只雄性大鼠随机分为五个组:对照组、模型对照组、美沙拉嗪组、酪酸梭菌组和美沙拉嗪和酪酸梭菌联合治疗组,每组10只。用酶联免疫吸附试验检测各组大鼠血清中炎性因子即IL-4、IL-6的含量。结果模型对照组与对照组比较,IL-4水平明显降低(P <0. 01),IL-6水平明显升高(P <0. 01)。活动期至缓解期美沙拉嗪组、酪酸梭菌组以及联合治疗组与模型对照组比较,IL-4水平显著升高,IL-6的水平显著下降,且差异均有统计学意义(P <0. 01)。而联合治疗组分别与美沙拉嗪组和酪酸梭菌组比较,血清IL-4和IL-6水平变化,有显著差异(P <0. 01)。结论美沙拉嗪和酪酸梭菌对溃疡性结肠炎都有显著的治疗效果,而联合治疗组效果更好,其作用机制可能与降低血清中IL-6的水平,升高血清IL-4的水平有关。
【Abstract】 Objective To explore the treatment effect of mesalasize combined with clostridium butyricum on ulcerative colitis in rats to find its possible mechanism of treating UC. Methods UC rat models,established by enteroclysis with 2,4,6-trinitrobenzene sulfonic acid( TNBS) and 50% ethanol solution,then 40 male rats were divided into four groups: control group,model groups,the mesalasize group,the clostridium butyricum group and ten in each group. Enzyme linked immunosorbent test each activity and remission of rats colon mucosa inflammatory factor namely the expression of IL-4 and IL-6. Results Model group compared with control group,IL-4 levels significantly decreased( P < 0. 01),IL-6 levels increased significantly( P < 0. 01). Activity to remission the mesalasize and clostridium butyricum treatment group compared with model group,IL-4 rising faster( mesalasize P < 0. 01,clostridium butyricum P < 0. 01),IL-6 cut speed is faster( mesalasize P < 0. 01,clostridium butyricum P < 0. 01).Conclusion The mesalasize and clostridium butyricum has significant therapeutic effect on UC,and the mesalasize group therapy effect is more prominent.
