壬基酚暴露对SD大鼠心肌组织的影响研究
Effects of nonylphenol exposure on myocardial tissue of SD rats
【摘要】 目的探讨壬基酚暴露对SD大鼠心肌组织的影响。方法选取健康雌性SD大鼠24只,按体重随机分为4组,每组6只。实验组分别给予25、50、100mg/kg壬基酚灌胃染毒,对照组给予植物油灌胃,每天1次,共42d。计算各组SD大鼠心脏脏器系数,采用分光光度法测定心肌组织Na+K+-腺嘌呤核苷三磷酸(ATP)酶、Ca2+Mg2+-ATP酶的活性,通过苏木精-伊红染色(HE染色)观察心肌组织病理变化。结果实验组SD大鼠体重、Na+-K+-ATP酶活性与对照组比较,差异均无统计学意义(P>0.05)。与对照组比较,实验组SD大鼠心脏质量、心脏系数、Ca2+Mg2+-ATP酶活性显著下降,差异均有统计学意义(P<0.05),且心脏质量、心脏系数与染毒剂量呈负相关(P<0.05)。病理组织学结果显示,对照组心肌组织纤维排列致密,结构清晰。25mg/kg染毒组未见病理形态改变,50mg/kg染毒组可见间质中有少量炎症细胞浸润,100mg/kg染毒组可见纤维排列比较疏松,部分心肌纤维结构破坏,细胞核消失,间质中炎症细胞浸润情况也较50mg/kg染毒组显著。结论壬基酚暴露可导致心肌组织炎性反应,使Ca2+Mg2+-ATP酶活性下降。
【Abstract】 Objective To investigate the effects of nonylphenol exposure on myocardial tissue in SD rats.Methods Twentyfour healthy female SD rats were randomly divided into 4 groups according to body weight,with 6 rats in each group.The experimental group was given 25,50,100 mg/kg nonylphenol by intragastric administration,and the control group was given vegetable oil by gavage once a day for 42 days.The cardiac organ coefficients of SD rats in each group were calculated.The activity of Na+K+-adenosine triphosphate(ATP)ase and Ca2+Mg2+-ATPase in myocardial tissue was determined by spectrophotometry.Pathological changes of myocardial tissue were observed by hematoxylin-eosin staining(HE staining).Results Compared with the control group,there was no significant difference in body weight,Na+-K+-ATPase activity of the SD rats in the experimental group(P>0.05).Compared with the control group,the heart mass,cardiac coefficient,Ca2+Mg2+-ATPase activity of the SD rats in the experimental group decreased significantly,and the differences were statistically significant(P<0.05).Cardiac mass and cardiac coefficient of SD rats in the experimental group were negatively correlated with the dose(P<0.05).Histopathological results showed that the myocardial fibers of the control group were densely arranged and the structure was clear.No pathological changes were observed in the 25 mg/kg group.In the 50 mg/kg group,a small amount of inflammatory cells infiltrated in the interstitium.In the 100 mg/kg group,the fiber arrangement was loose,some myocardial fiber structure was destroyed,the nucleus disappeared,and the inflammatory cell infiltration in the interstitial was also significantly higher than that in the 50 mg/kg group.Conclusion Exposure to nonylphenol can cause an inflammatory response in myocardial tissue,and make Ca2+Mg2+-ATPase activity decrease.
