超声触发载多西紫杉醇纳泡对兔VX2乳腺癌肿瘤模型的靶向抗肿瘤作用及监控

【摘要】 目的研究超声触发载多西紫杉醇纳泡(LDLN)对兔VX2乳腺癌肿瘤模型的靶向抗肿瘤作用及监控。方法采用瘤块包埋法建立兔VX2肿瘤模型,将60只荷瘤兔随机平均分为6组:对照组(C组)、LNLD组、单纯药物组(Doc组)、药物联合超声组(Doc+US组)、单纯纳泡联合超声组(PLM+US组)、LDLN联合超声组(LNLD+US组),给予各组不同的药物注射及处理方法。各组治疗前后均行超声检查观察肿瘤的生长变化情况,计算并比较各组的肿瘤体积及体积抑瘤率(TIR)。光镜下观察各处理组对兔VX2乳腺瘤组织细胞形态学的影响。结果在一定强度的超声作用下,LNLD+US组的TIR高于其他组,差异均有统计学意义(P<0.05);各给药组瘤组织内出现不同程度坏死区。结论超声触发LDLN对兔VX2乳腺癌肿瘤模型具有靶向抗肿瘤及监控的双重作用。

【Abstract】 Objective To study the antitumor effect and monitoring of ultrasound triggered docetaxel loaded nanoparticles(LDLN)on rabbit VX2 breast cancer model.Methods The tumor block embedding method was used to establish the rabbit VX2 tumor model.Sixty tumor-loading rabbits were randomly divided into 6 groups:control group(group C),LNLD group,pure drug group(Doc group),drug combined with ultrasound group(Doc+US group),pure nanobubble combined ultrasound group(PLM+US group),LDLN combined with ultrasound group(LNLD+US group).Various groups were given different drug injections and treatment methods.The growth and changes of tumor growth were observed by ultrasonography before and after treatment in each group,the tumor volume and volume tumor suppression rate(TIR)were calculated and compared among the groups.The effect of each treatment group on the histological morphology of VX2 mammary tumor in rabbits was observed under light microscope.Results After certain intensity of ultrasound,TIR of the LNLD+US group was higher than that of the other groups,the differences were statistically significant(P<0.05).Different degrees of necrosis area appeared in the tumor tissues of each treatment group.Conclusion Ultrasound-triggered LDLN has a dual role of targeted anti-tumor and surveillance in rabbit VX2 breast cancer tumor model
