地氯雷他定治疗新生大鼠低温肺出血疗效研究
Study on effect of desloratadine in treating hypothermic lung hemorrhage in neonatal rat
【摘要】 目的探讨第3代抗组胺药物——地氯雷他定对新生大鼠低温肺出血的治疗作用。方法 2012年2月将60只SD级新生大鼠按照完全随机法分为常温对照组(C组,20只)和低温建模组(40只);将40只低温建模组大鼠按照完全随机法分为低温对照组(L组)和地氯雷他定组(D组),每组20只。给药后于1、2、3、4 h时间点处死各组大鼠(每个时间点5只),采集标本后观察各组大鼠肺组织苏木精-伊红染色病理学改变,检测肺组织湿重/干重(W/D)比值、组胺含量,以及血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平。结果低温建模组大鼠肺组织病理学改变情况、肺组织W/D比值与C组比较,差异均有统计学意义(P <0. 05); D组大鼠肺组织病理学改变情况、肺组织W/D比值与L组比较,差异均有统计学意义(P <0. 05)。L、D组大鼠各时间点肺组织组胺水平及血清TNF-α、IL-6水平均明显高于C组,D组各时间点大鼠肺组织组胺水平及血清TNF-α、IL-6水平均明显低于L组,差异均有统计学意义(P <0. 05)。结论地氯雷他定对新生大鼠肺部出血性损伤具有保护作用,对新生大鼠低温肺出血具有一定的治疗作用。
【Abstract】 Objective To study the therapeutic effects of third generation antihistamine drug desloratadine on lung hemorrhage induced by hypothermia in neonatal rat. Methods Sixty Sprague-Dawley( SD) grade neonatal rats were randomly divided into the normal temperature control group( group C,20 cases) and hypothermia model group( 40 cases); the hypothermia model group was randomly re-divided into the hypothermia control group( group L) and desloratadine group( group D),20 cases in each group. The rats in each group were killed at the timepoints of 1,2,3,4 h after medication( 5 cases at each time point). The HE staining pathogenic changes of lung tissue,lung wet/dry( W/D) weight ratio,histamine content and levels of TNF-α and IL-6 in serum were observed in each group. Results The lung tissue pathologic changes and lung tissue W/D ratio had statistical differences between the hypothermia model group and group C( P < 0. 05);the lung tissue pathologic changes and lung tissue W/D ratio had statistical differences between the group D and L( P < 0. 05). The histamine level of lung tissue and serum TNF-α and IL-6 levels at each time point in the group L and D were significantly higher than those in the group C( P < 0. 05),while the histamine level of lung tissue and serum TNF-α and IL-6 levels at each time point in the group D were significantly lower than those in the group L,and the differences were statistically significant( P < 0. 05). Conclusion Desloratadine has the protective effect on lung hemorrhage injury in neonatal rat and has a certain treatment effect on hypothermic lung hemorrhage in neonatal rat.
