溺死大鼠肺组织和水样中硅藻检验的定量对比研究
Quantitative Comparison of Diatoms in Both the Lungs of Rats Died from Drowning and Drowning Media
【摘要】 目的研究大鼠肺组织中硅藻与水样中硅藻数量的比值判断死亡原因的应用价值和判断标准。方法取36只成年SD大鼠随机分入双蒸水组、高浓度硅藻组、低浓度硅藻组三个大组中(各12只),各大组再分为溺死组和死后抛尸组,每组(n=6)应用微波消解-真空抽滤法,对肺组织中硅藻含量与水样中硅藻浓度进行分析。结果动物实验证实大鼠溺水死亡后,肺组织中检出的硅藻量与水样中硅藻浓度有关,溺死大鼠的肺组织与水样硅藻含量比平均值为0.47±0.07且其95%置信区间为(0.32,0.61)。结论肺组织硅藻检验的定量分析结果可作为推断水中尸体死亡原因的辅助依据,本研究提供了一种基于肺组织与水样硅藻含量比的溺死判断标准。
【Abstract】 Death cause of corpse in water plays an important part for forensic medical examination, yet difficult to diagnose as "true drowning" based solely on postmortem phenomenon. Currently, diatom testing, albeit considered as the"golden standard" for drowning diagnosis, remains controversial. This study aimed to investigate the value of quantitative diatom from deceased rats’ lung tissues against the involved water samples, to discover and diagnose the death cause of corpses out of drowning. Thirty-six SD rats were randomly divided into 6 groups, conducting with different experimental treatments to their life end. All the deceased rats’ lung tissues were collected, having diatoms recovered from each of them through microwave digestion-vacuum filtration. The comparison indicates that the lung-tissue-carried diatom quantity is of positive correlation with the diatom density of related water samples. The average ratio(diatom counts in lung tissues of drowned rats to the ones in related water sample) is 0.47±0.07, with the 95% CI as 0.32, 0.61. Accordingly, quantitative diatom in lung tissue can be an auxiliary reference for estimating the death cause of submerged cadaver, thus offering a standard for drowning diagnosis by the ratio of diatom counts in the deceased’s lung tissue to the relevant water sample.
