基于响应面分析法优化甘蔗梢提取物中黄酮分离工艺研究

【摘要】 采用响应面分析法研究大孔吸附树脂分离甘蔗梢提取物中黄酮的最佳工艺条件.在单因素试验的基础上,通过旋转中心组合试验设计原理,进行三因素三水平试验,以上样p H值、流速和洗脱体积为考察指标,建立黄酮含量的多元二次回归方程,得到了优化组合条件.结果表明,不同分离条件对黄酮含量影响由大到小的顺序为:洗脱体积> p H值>流速,同时得到较优的分离工艺条件为:甘蔗梢提取物用蒸馏水溶解后调节p H值为6. 71,然后以提取物质量(干重)与大孔吸附树脂比为1:20上样,选择D141型大孔吸附树脂为分离介质,运用柱层析分离,以体积分数95%的乙醇溶液为洗脱液,控制流速为2. 50 BV/h,洗脱体积为4. 00 BV时,有较好的分离效果.此条件下黄酮收率为193 mg/kg,与模型预测值接近.

【Abstract】 The purpose of the current study was the optimization of the process for separation of flavonoids from sugarcane tip extracts using response surface methodology. A central composite design was used to investigate the best conditions for separation of flavonoids. In the separation procedure,the three independent variables were p H value,flow rate,and volume times of elution volume to column volume. According to the flavonoids content as response value,a multivariable quadratic regression equation was developed. The results showed that the sequence affecting the content of flavonoids was volume times of elution volume to column volume > p H value > flow rate. And the optimal separation conditions for flavonoids from sugarcane tips were as follows: D141-type resin was chosen as the best macroporous adsorptive resin in the separation of flavonoids from sugarcane tips,pH 6. 71,weight ratio of resin to crude sample of 20,adding 95% ethanol four times the column volume for flavonoids with a flow rate of 2. 50 BV/h. Under these conditions,the yield of flavonoids was up to 193 mg/kg,which approached to the predicted value.
