沉默环状RNA hsacirc0011021表达对宫颈癌细胞增殖迁移及侵袭的影响

【摘要】 目的探讨环状RNA(circRNA)hsacirc0011021在宫颈癌组织和细胞株中的表达变化及其对宫颈癌细胞增殖、迁移和侵袭能力的影响。方法选取河南科技大学第一附属医院就诊的15例宫颈癌患者,取15对宫颈癌组织和癌旁组织。qRT-PCR实验检测hsacirc0011021在宫颈癌组织和宫颈癌癌旁组织(Hela和SiHa)细胞中的相对表达水平;CCK-8和克隆形成实验检测沉默hsacirc0011021对Hela细胞活性和克隆形成能力的影响;划痕愈合实验和Transwell侵袭实验检测沉默hsacirc0011021对Hela细胞迁移及侵袭能力的影响。结果 hsacirc0011021在宫颈癌组织的相对表达水平明显高于癌旁组织(P<0.05);hsacirc0011021在Hela和SiHa细胞中的相对表达水平明显高于人永生化表皮细胞HaCaT(P<0.05)。在Hela细胞沉默hsacirc0011021,Hela细胞的活性和克隆形成能力显著降低,迁移距离明显减少,侵袭能力也明显下降(P<0.05)。结论 hsacirc0011021在宫颈癌组织和宫颈癌细胞(Hela和SiHa)中的表达水平明显升高;沉默hsacirc0011021显著抑制宫颈癌Hela细胞的增殖、迁移和侵袭。

【Abstract】 Objective To explore the expression of the circular RNA(circRNA) hsacirc0011021 in cervical cancer tissues and cell lines, and study the effect of hsacirc0011021 knockdown on the proliferation, migration and invasion of cervical cancer cells. Methods The expression levels of hsacirc0011021 in cervical cancer tissues and cervical cancer cell lines(Hela and SiHa) were determined by quantitative real-time PCR(qRT-PCR). The effect of hsacirc0011021 on the viability and proliferation of cervical cancer cells, CCK-8 and colony formation assays were performed in Hela cells were observed. Transwell migration assay and wound scratch assay were carried out to evaluate the effect of hsacirc0011021 on the migration and invasion ability of Hela cells.Results The expression level of hsacirc0011021 in cervical cancer tissues and Hela and SiHa cells was higher than that in adjacent normal tissues and human immortal keratinocyte line(HaCaT). Knockdown of hsacirc0011021 obviously inhibited the proliferation, migration and invasion of Hela cells. Conclusion Knockdown of hsacirc0011021 remarkably suppresses the proliferation, migration and invasion of cervical cancer cells.
