培养基优化对菊苣茎乙醇提取物抑制粪肠球菌增殖效果的影响

【摘要】 目的研究菊苣茎乙醇提取物抑制粪肠球菌增殖的效果及其机制。方法采用星点设计-响应面法优化粪肠球菌的培养条件;用生长曲线法测定菊苣茎乙醇提取物处理粪肠球菌的吸光度值A;用抑菌圈直径测定菊苣茎乙醇提取物抑制粪肠球菌的效果;通过测定细菌胞外碱性磷酸酶(AKP)含量变化分析菊苣茎乙醇提取物损伤粪肠球菌细胞壁情况。结果粪肠球菌的优化培养条件为:葡萄糖浓度0.02 mol·L-1,甘油浓度0.02 mol·L-1,培养时间331.19 min;菊苣茎乙醇提取物对粪肠球菌具有明显的抑制增殖作用。结论优化后的培养条件可促进粪肠球菌增殖,菊苣茎乙醇提取物通过增加粪肠球菌细胞膜通透性发挥抑菌效果。

【Abstract】 Objective To study the effect of ethanol extract of chicory stem on the proliferation of Enterococcus faecalis and its mechanism.Methods Central composite design-response surface methodology was used to optimize the culture conditions of Enterococcus faecalis.Growth curve method was used to determine the absorbance A value of Enterococcus faecalis treated with ethanol extract of chicory stem.Inhibitory effect of ethanol extract of chicory stem on Enterococcus faecalis was evaluated by inhibition zone method.The effects of ethanol extract from chicory stem on the cell wall of fecal enterococcus was analyzed by measuring the changes in the content of bacterial extracellular alkaline phosphatase(AKP).Results The optimal culture conditions of Enterococcus faecalis were glucose 0.02 mol·L-1,glycerol 0.02 mol·L-1,and culture time 331.19 min;the ethanol extract of chicory stem had obvious inhibitory effect on Enterococcus faecalis.Conclusion The optimized culture conditions can promote the proliferation of Enterococcus faecalis.The ethanol extract of chicory stem exerts the antibacterial effect by increasing the cell membrane permeability of Enterococcus faecalis.
