UPLC-MS法对盐肤木果实及其油脂中漆酚成分的分析
UPLC-MS Analysis of Urushiol in the Fruit and Oil of Rhus chinensis
【摘要】 盐肤木果实中特别是其果油中是否含有致敏性成分——漆酚,一直是盐肤木果油作为食用油脂的安全隐患之一。以饱和漆酚为标准对照,采用UPLC-MS分析法,对3种盐肤木果油等4个样品中的饱和漆酚,进行了定性、定量分析。MS定性分析表明,饱和漆酚标准品中的漆酚为其分子中R为C15的饱和烃链,其分子式为C21H36O2,再以二级质谱中质荷比为122.037的分子碎片离子流强度作为定量分析指标,以标准曲线法对4个样品中饱和漆酚进行了定量分析;再通过一级质谱的分析,对可能的7种不同结构的不饱和漆酚在4个样品中的含量进行了初步分析。结论:4个样品中均含有饱和漆酚,其含量在27.5—57.5μg/L之间,盐肤木果油中总漆酚含量应为165—345μg/L左右。
【Abstract】 Whether urushiol, the allergenic ingredient, is contained in the fruit of Rhus chinensis, especially in the fruit oil, has always been one of the hidden safety risks of Rhus chinensis fruit oil as edible oil. The saturated urushiol supplied by Xi’an institute of raw lacquer was used as the standard reference, the method was analyzed by UPLC-MS, the saturated urushiol in 4 samples, including the fruit oil by 3 different extraction methods and the other was ethanol extract from Rhus chinensis fruit, was analyzed qualitatively and quantitatively. UPLC-MS analytical results showed that urushiol in the saturated urushiol was the saturated hydrocarbon chain with R of C15 in its molecule, its molecular formula is C21H36O2. Furthermore, the ion flow intensity of molecular debris with m/z=122.037 was selected for a quantitative analysis index, and the saturated urushiol in 4 samples was quantitatively analyzed by standard curve method. The results showed that the content of urushiol in 4 samples ranged from 27.5 mg/L to 57.5 mg/L. Finally, the contents of 7 different structures in unsaturated urushiol within 4 samples were evaluated by primary mass spectrometry.
