乏氧环境下TIGAR线粒体定位介导食管癌乏氧辐射抵抗
The Study of the Effect of TIGAR in Hypoxia-induced Esophageal Cancer Radioresistance
【摘要】 目的研究乏氧环境下TIGAR线粒体定位变化对乏氧人食管癌细胞放射敏感性的影响。方法通过western-blot检测不同食管癌细胞株的TIGAR线粒体定位水平及乏氧对TIGAR线粒体定位水平的影响;通过克隆形成实验检测调控乏氧环境下TIGAR线粒体定位水平对于食管癌细胞辐射敏感性的影响。结果 (1)不同食管鳞癌细胞(ECA109、KYSE410、KYSE450)内TIGAR的线粒体定位水平不同,其中KYSE410细胞TIGAR在线粒体中的表达最高,KYSE450的表达最低。(2)在乏氧培养条件下,食管鳞癌细胞中TIGAR在线粒体中的表达升高。(3)在干扰KYSE410细胞TIGAR表达后,细胞在乏氧条件下辐射敏感性增加;而在KYSE450细胞过表达TIGAR后,细胞在乏氧条件下辐射敏感性减弱。结论乏氧可以诱导食管鳞癌细胞中的TIGAR线粒体定位,从而介导食管癌乏氧辐射抵抗。
【Abstract】 Objective To study the effect of TIGAR in hypoxia-induced esophageal cancer radioresistance. Methods The expression level of TIGAR in the mitochondria of esophageal cell lines was detected by Western-blot. Colony formation assay was performed to study the effect of TIGAR in radiosensitivity of hypoxic esophageal cancer cells. Results KYSE410 cells had the highest expression of TIGAR and KYSE450 had the lowest. Under hypoxic culture conditions, the expression of TIGAR in mitochondria was elevated. 3. After knocking down the TIGAR expression, the radiosensitivity of hypoxic esophageal cancer cells was increased. After overexpressing TIGAR, the radiosensitivity of hypoxic esophageal cancer cells was decreased. Conclusion Hypoxia can induce TIGAR mitochondria localizatio, which can mediate the hypoxia-induced esophageal cancer radioresistance.
