酸枣仁对酒精戒断焦虑征的治疗作用

【摘要】 目的研究酸枣仁对酒精戒断焦虑征的治疗作用,并探讨其作用机制。方法选取健康的SD大鼠进行随机分组,腹腔内注射酒精28天,制备酒精成瘾模型,灌胃不同制备方法及不同剂量酸枣仁3天进行治疗,最后一次灌胃1h后,高架十字迷宫测其焦虑行为,HPLC测大鼠海马中的去甲肾上腺素(NE)的含量。结果高架十字迷宫检测结果显示,相比较空白组,酒精戒断焦虑征大鼠停留在开放臂的时间及次数显著减少(P<0.05);相比较模型组,灌胃大剂量的炒制酸枣仁醇提物后其次数和时间显著增加(P<0.01),灌胃小剂量炒制的酸枣仁醇提物后能增加停留的时间和次数(P<0.05),灌胃大、小剂量的酸枣仁醇提取物对焦虑症状的作用效果不显著(P>0.05);HPLC检测显示炒制酸枣仁醇提物有降低海马中去甲肾上腺素(NE)含量的作用(P<0.05),酸枣仁醇提物的作用效果不明显(P>0.05)。结论灌胃炒制的酸枣仁醇提取物可减轻大鼠酒精戒断后出现的焦虑行为,抗焦虑的作用机制可能与海马中NE的含量降低有关。

【Abstract】 Objective In the present study, we investigated the effect of Semen Ziziphi Spinosae on anxiety-like behavior and the levels of norepinephrine in Hippocampus during ethanol withdrawal in rats. Methods Healthy SD rats were randomly divided. In addition to the blank group, the remaining groups were intraperitoneally injected with 20%(w/v) alcohol 1.5 ml/100 g per day for 28 days to prepare an alcoholic addiction model, and then the drug was given once a day for 3 days. One hour after the last administration,the anxiety behavior was measured by the elevated cross maze, and the content of norepinephrine in the hippocampus was determined by HPLC. Results Rats undergoing ethanol withdrawal exhibited substantial anxiety-like behavior, which was characterized by the decrease in time spent in the open arms of the elevated plus maze,which were greatly attenuated by doses of stir-frying Semen Ziziphi Spinosae.The high performance liquid chromatography analysis revealed that ethanol withdrawal significantly increased norepinephrine levels in the hippocampus. Similar to the results from the elevated plus maze test, the stir-frying Semen Ziziphi Spinosae significantly inhibited the elevation of norepinephrine. Conclusions These results suggest that Semen Ziziphi Spinosaee attenuate anxiety-like behavior induced by ethanol withdrawal through modulation of the hypothalamic norepinephrine system in the brain.
