STAT3信号通路在多发性骨髓瘤中的作用及相关的药物治疗进展

【摘要】 信号转导和转录激活信号通路,即STATs信号通路,在不同的生物学过程中发挥着重要作用,包括免疫反应,造血过程以及神经发生等。在STAT成员中,STAT3在细胞增殖、存活与分化中发挥着至关重要的作用。虽然在生理条件下,STAT3的激活是短暂的,但在高比例的实体和恶性血液瘤中,STAT3会持续激活,从而促进肿瘤的发生发展与恶性转化。多发性骨髓瘤是一种血液学恶性肿瘤,其特征是骨髓中血浆细胞的恶性克隆增殖。STAT3信号通路在多发性骨髓瘤中持续激活,促进了其增殖分化,也是多发性骨髓瘤难以完全治愈的原因之一。在本文中,我们总结了有关STAT3相互作用因子在多发性骨髓瘤中作用的数据,并讨论其发生作用的具体机制以及靶向该通路对于多发性骨髓瘤的相关治疗进展。

【Abstract】 Signal transduction and transcriptional activation signaling pathway, namely STATs signaling pathway, plays an important role in different biological processes, including immune response, hematopoietic process and neurogenesis. STAT3 plays an important role in cell proliferation, survival and differentiation among STAT members. Although the activation of STAT3 is transient under physiological conditions, it will continue to activate in a high proportion of solid and malignant hematomas, thus promoting the occurrence, development and malignant transformation of tumors. Multiple myeloma is a hematological malignancy characterized by malignant clonal proliferation of plasma cells in bone marrow. STAT3 signaling pathway continues to activate in multiple myeloma,which promotes its proliferation and differentiation, and is also one of the reasons why multiple myeloma is difficult to cure completely.In this article, we summarize the data on the role of STAT3 interacting factors in multiple myeloma, and discuss the specific mechanism of its action and the progress of targeted therapy for multiple myeloma.
