HPLC法测定大鼠血浆中胆红素的含量
HPLC determination of bilirubin in rat plasma
【摘要】 目的建立反相HPLC法测定血浆中胆红素含量。方法色谱柱:C18柱(4. 6 mm×250 mm,5μm),流动相:甲醇-1%醋酸溶液(95∶5);流速:1. 0 ml·min-1;检测波长:455 nm,柱温为室温。结果血浆中胆红素在17. 5~280. 0μg·ml-1的浓度范围内,A=73131C-999834(r=0. 9965),线性关系良好;胆红素平均回收率(n=9)分别是108. 15%,114. 73%,102. 46%,RSD值为1. 5%。结论该含量测定方法准确,快速,有效,可用于血浆中胆红素的含量测定。
【Abstract】 Objective: To establish a method for determination of bilirubin in rat plasma by high performance liquid chromatography( HPLC). Methods: Bilirubin was detected at the wavelength of 455 nm with C18 column( 250 mm × 4. 6 mm,5μm). The mobile phase was methyl alcohol-1% ethanoic acid( 95∶ 5) and flow rate was 1. 0 ml·min-1; the column temperature was room temperature. Results: The linear range of bilirubin was within 17. 5 ~ 280. 0 μg·ml-1,A = 73131 C-999834( r = 0. 9965). The average recovery rate( n = 9) was 108. 15%,114. 73%,101. 88%,RSD value was 1. 5%. Conclusion:The method is accurate,rapid,simple and can be used in quality control for bilirubin.
