UPLC-ESI-MS/MS法测定人参、红参和西洋参中多种皂苷含量

【摘要】 建立了人参、红参和西洋参中32种皂苷的超高效液相色谱-电喷雾-串联四极杆质谱检测方法。电离模式:负离子(ES-);检测方式:多反应监测(MRM);洗脱液:乙腈-水(均含2.5 mmol/L甲酸);洗脱方式:梯度;色谱柱:ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 m);柱温:45℃。分析用时29 min。各皂苷的定量限为0.001~0.013 g/kg,线性校准曲线回归方程的回归系数均在0.995以上。该方法简便、可靠,适合人参、红参、西洋参中多种皂苷的同时快速测定。

【Abstract】 The aim of the present study was to developan ultra-high performance liquid chromatography-electrospray-tandem quadrupole mass spectrometrymethod for the determination of 32 saponins in ginseng, red ginseng and american ginseng. Ionization mode: negative ion(ES-); detection mode: multiple reaction monitoring;eluent: acetonitrile-water(containing 2.5 mmol/L formic acid); elution method: gradient; column: ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 m); column temperature:45 ℃.The analysis time:29 minutes.The quantitative limits of saponins ranged from 0.001-0.013 g/kg, and the regression coefficients of linear calibration curve regression equation were all above 0.995.The method is simple, reliable and suitable for the simultaneous determination of various saponins in ginseng, red ginseng and american ginseng.
