砷对星形胶质细胞上EAAT1、EAAT2和xCT蛋白水平的影响

【摘要】 目的探讨急性砷暴露对星形胶质细胞(AC)上EAAT1、EAAT2和xCT蛋白水平的影响。方法 AC在不同浓度和不同时间砷暴露后,利用高效液相色谱法和微量滴定板法测定AC内GSH的含量以及细胞活力,使用Western Blot法检测AC上EAAT1、EAAT2和xCT蛋白表达水平。结果与对照组比较,随着染砷浓度的增加和时间的延长,产生了明显的细胞毒性,AC内GSH水平表现出应激性增加,AC膜上EAAT1、EAAT2蛋白表达水平明显升高,xCT蛋白表达水平明显降低,差异有统计学意义(P<0.05)。结论急性砷暴露可产生明显细胞毒性,砷可促进AC膜上xCT蛋白表达,抑制EAAT1、EAAT2蛋白的表达。

【Abstract】 Objective To investigate the effect of acute arsenic exposure on the protein levels of EAAT1, EAAT2 and xCT in Astrocytes(AC). Methods After arsenic exposure at different concentrations and times, GSH content and cell viability in AC were tested by high performance liquid chromatography and microfiltration plate. Protein levels of EAAT1, EAAT2 and xCT of AC were tested by Western Blot. Results Compared with the control group, with the increase of arsenic concentration and the prolongation of time, obvious cytotoxicity was produced; the GSH in AC showed hyperirritability; the protein expression levels of EAAT1 and EAAT2 of AC were significantly increased, and the protein level of xCT were significantly decreased. Conclusion Acute arsenic exposure can produce obviously cytotoxicity. Arsenic can promote the expression of xCT protein of AC and inhibit the expression levels of EAAT1 and EAAT2 proteins.
