着床前遗传学筛查周期中不同受精来源囊胚发育速度与染色体整倍性分析

【摘要】 目的为了提高着床前遗传学筛查(PGT)周期患者体外授精-胚胎移植(IVF-ET)成功率和胚胎利用率,通过单核苷酸多态性-微阵列比较基因组杂交技术(array-SNP)分析PGT患者的2PN、0PN和1PN来源的胚胎染色体的整倍性,从而优化移植策略。方法回顾性分析2013—2018年在我中心行PGT助孕,其2PN、1PN和0PN来源的胚胎均有囊胚形成的患者,比较不同发育速度的2PN、0PN和1PN来源的活检囊胚的染色体整倍性。结果比较第5天不同受精来源的活检囊胚染色体整倍体率差异无统计学意义(P>0.05);第6天囊胚中,0PN来源的活检囊胚染色体整倍体率要低于2PN和1PN来源的活检囊胚整倍体率,差异有统计学意义(P<0.05);第5天和第6天的囊胚相比,不同受精来源的囊胚染色体整倍体率差异无统计学意义(P>0.05)。结论对于PGT周期患者,0PN以及1PN来源的胚胎培养后形成囊胚,通过芯片技术检测其染色体正常的囊胚仍占一定比例,可作为移植胚胎。不同发育速度的囊胚,其染色体的整倍体率差异并无统计学意义。

【Abstract】 Objective The aim to analyze the chromosome ploidy of 2 PN,0 PN and 1 PN-derived embryos in patients with preimplantation genetic testing(PGT) by array-SNP,in order to optimize the transplantation strategy,thereby improving the pregnancy rate and embryo utilization rate of IVF-ET. Methods In this retrospective study,we compared the chromosome ploidy of biopsy-derived blastocysts from 2 PN,0 PN and 1 PN with different development rates in the patients who had blastocyst formation in 2 PN,1 PN and 0 PN-derived embryos in our center from 2013 to 2018.Results There was no significant difference in chromosome ploidy rate between the blastocysts from different fertilization sources on day 5(P> 0.05). On day 6,the chromosome ploidy rate of 0 PN-derived blastocysts was significantly lower than that of 2 PN and 1 PN-derived blastocysts from biopsy(P <0.05). There was no significant difference in the chromosome ploidy rate of blastocysts from different sources between day 5 and day 6(P>0.05). Conclusion For the patients with PGT cycle,a certain proportion of blastocysts,formed from 0 PN and 1 PN-derived embryos,have normal chromosomes as determined by the chip technology,which could be used for embryo transplantation.
