靶向ADAM17基因siRNA对食管癌EC109细胞增殖及迁移能力的影响

【摘要】 目的探讨靶向ADAM17基因的小干扰RNA(siRNA)对EC109食管癌细胞增殖及迁移能力的影响。方法将靶向人ADAM17基因的siRNA分别标记为si257,si258及si259三个序列,转染入EC-109人食管癌细胞;采用Realtime PCR法检测各组癌细胞ADAM17 mRNA的含量; Cell Counting Kit-8(CCK8)实验用于检测ADAM17沉默后食管癌细胞EC109的细胞增殖能力的变化; Transwell技术检测siRNA干扰后EC109迁移能力的变化。结果三种序列均能够有效抑制EC109中ADAM17的表达,显示有效的基因沉默效果。si259对EC109的增殖能力具有显著抑制作用。未观察到si257对于EC109增殖的抑制作用。迁移实验24 h后,未观察到siRNA的干扰对于EC109迁移有任何影响。结论 siRNA沉默ADAM17基因能够对食管癌细胞增殖产生一定的影响,但未观察到siRNA的干扰对于食管癌细胞迁移的影响,提示ADMA17基因通过某种通道参与了食管癌细胞的增殖活动,并且在食管癌侵袭、转移的活动中可能需要体内外某种介质的信号传导。

【Abstract】 Objective To investigate the effect of small interference RNA( siRNA) targeting ADAM17 gene on the proliferation and migration of EC109 esophageal cancer cells. Methods The ADAM17-siRNA was marked with three sequences including si257,si258 and si259,respectively,and transferred into EC-109 human esophageal cancer cells,and the content of ADAM17 mRNA in each group was detected by Realtime PCR. Cell Counting Kit-8( CCK8) assay was used to detect the changes of cell proliferation of esophageal cancer cell line EC109 after ADAM17 silencing,and Transwell technique was used to detect the change of EC109 migration ability after siRNA interference. Results All three sequences could effectively inhibit the expression of ADAM17 in EC109,showing an effective gene silencing effect. Si259 had a significant inhibitory effect on the proliferation of EC109. The inhibitory effect of si257 on the proliferation of EC109 was not observed. After 24 hours of migration experiment,no effect of siRNA interference on EC109 migration was observed. Conclusion SiRNA silencing ADAM17 gene can have some effect on the proliferation of esophageal cancer cells,but no effect of siRNA interference on the migration of esophageal cancer cells has been observed,suggesting that ADMA17 gene is involved in the proliferation of esophageal cancer cells through a certain channel,and the signal transduction of some medium may be required in the invasion and metastasis of esophageal cancer.
