芝麻素对冠心病大鼠脂代谢、血管内皮功能损伤及VCAM-1水平的影响
Effects of sesamin on lipid metabolism,vascular endothelial function damage and VCAM-1 level in rats with coronary heart disease
【摘要】 目的观察不同剂量芝麻素对冠心病大鼠脂代谢、血管内皮功能、血管细胞黏附分子-1(VCAM-1)水平的影响。方法根据治疗药物及浓度不同,分为芝麻素低剂量组(L组)、中剂量组(M组)、高剂量组(H组,另设基础对照组(C组),冠心病模型组(CHD组)。C组大鼠仅给予普通基础饲料喂养,其他4组大鼠建立大鼠冠心病模型,给予高脂饲料饲养。C组和CHD组采用等体积生理盐水灌胃,L组、M组和H组依次采用芝麻素芝麻素40 mg/kg、80 mg/kg、160 mg/kg灌胃。观察治疗6周后大鼠脂代谢、血管内皮功能、VCAM-1变化。结果 CHD组大鼠总胆固醇(TC)含量显著高于C组,CHD组TC含量显著高于L组、M组、H组,随着芝麻素剂量的增加,TC下降程度也显著增加(P<0. 05)。治疗后各组甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)含量差异无统计学意义(P> 0. 05)。冠心病大鼠血管最大舒张程度由(99. 82±3. 06)%降低至高剂量组的(52. 06±5. 15)%。随着芝麻素使用剂量的增加,血管最大舒张程度显著降低,差异均有统计学意义(P <0. 05)。而给予10-8~10-4mol/L累积浓度的酸化Na NO2,各组间血管张力无显著差异(P> 0. 05)。CHD组、L组、M组、H组VCAM-1表达水平均显著高于C组(P <0. 05),随着芝麻素使用剂量的增加,VCAM-1表达量显著下降,组间比较差异有统计学意义(P <0. 05)。结论芝麻素可改善冠心病大鼠脂代谢、血管内皮功能,降低VCAM-1水平,随着治疗剂量的增加,上述改善效果更明显。
【Abstract】 Objective To observe the effects of different doses of sesamin on lipid metabolism,vascular endothelial function and VCAM-1 levels in rats with coronary heart disease. Methods According to different therapeutic drugs and concentrations,it was divided into low dose group of sesamin( L group),middle dose group( M group),high dose group( H group),a basic diet control group( group C) and a coronary heart disease model group( CHD group). Group C was only fed with normal basal diet,and the other 4 groups of rats were established with a model of coronary heart disease in rats,and fed with high-fat diet. Group C and CHD were intragastrically administered with equal volume of normal saline. Groups L,M and H were administered with sesamin 40 mg/kg,80 mg/kg,160 mg/kg intraperitoneal injection. Lipid metabolism,vascular endothelial function and VCAM-1 changes were observed in rats after 6 weeks of treatment. Results The TC content in CHD group was significantly higher than that in C group. The TC content in CHD group was significantly higher than that in L group,M group and H group. The dose of TC decreased significantly with the increase of dose( P < 0. 05). There was no significant difference in the levels of TG and HDL-C between the groups after treatment( P > 0. 05). The maximum relaxation of blood vessels in coronary heart disease rats decreased from( 99. 82 ± 3. 06) %to( 52. 06 ± 5. 15) % in the high dose group. With the increase of the dose of sesamin,the maximum relaxation of blood vessels was significantly decreased,and the difference was statistically significant( P < 0. 05). The acidified Na NO2 with a cumulative concentration of 10-8~ 10-4 mol/L was not significantly different between the groups( P > 0. 05). The expression levels of VCAM-1 in CHD group,L group,M group and H group were significantly higher than those in group C( P < 0. 05). With the increase of sesamin dosage,the expression of VCAM-1 was significantly decreased,and the difference between the groups was statistically significant( P < 0. 05). Conclusion Sesamin can improve lipid metabolism,vascular endothelial function and decrease VCAM-1 in rats with coronary heart disease. As the therapeutic dose increases,the above improvement effect is more pronounced.
