【摘要】 目的观察病毒性心肌炎患者外周血CD8~+T细胞自噬水平变化。方法回顾性分析天门市第一人民医院收治的80例病毒性心肌炎患者,作为观察组,40例健康体检者作为对照组。取患者外周血5 ml,以流式细胞术检测外周血CD8~+T细胞、效应性T细胞、记忆性T细胞百分比。流式分选CD8~+T细胞,收集CD8~+T细胞RNA及蛋白,检测自噬相关基因MAP1LC3、SQSTM1蛋白及mRNA表达情况。结果观察组CD8~+T淋巴细胞数、CD8效应T细胞、记忆性T细胞比率均显著高于对照组,差异有统计学意义(P <0.05)。观察组外周血CD8~+T细胞凋亡率显著低于对照组(P <0.05)。两组外周血CD8~+T细胞中MAP1LC3蛋白、mRNA表达量差异均无统计学意义(P> 0.05)。观察组外周血CD8~+T细胞中SQSTM1蛋白、mRNA表达水平均显著高于对照组(P <0.05)。结论病毒性心肌炎患者外周血CD8~+T细胞自噬水平增加,自噬体及自噬底物均显著增加,可能参与病毒性心肌炎的发病。更多还原

【Abstract】 Objective To observe the changes of autophagy level of peripheral blood CD8~+T cells in patients with viral myocarditis.Methods Eighty patients with viral myocarditis were selected as observation group,and 40 healthy examiners were used as control group.The5 mL peripheral blood of patients was taken,and the percentages of peripheral blood CD8~+T cells,effector T cells and memory T cells were detected by flow cytometry.CD8~+T cells were given fluorescence-activated cell sorting,and the RNA and protein of CD8~+T cells were collected,and the expression levels of autophagy-related genes MAP1 LC3 and SQSTM1 were detected.Results The ratios of CD8~+T lymphocytes,CD8 effector T cells and memory T cells in observation group were significantly higher than those in control group(P < 0.05).The apoptosis rate of peripheral blood CD8~+T cells in observation group was significantly lower than that in control group(P < 0.05).There was no significant difference in the expression of MAP1 LC3 mRNA in peripheral blood CD8~+T cells between the two groups(P > 0.05).The mRNA expression level of SQSTM1 in peripheral blood CD8~+T cells in observation group was significantly higher than that in control group(P < 0.05).Conclusion The autophagy level of peripheral blood CD8~+T cells is increased significantly in patients with viral myocarditis,and autophagosomes and autophagy substrates are increased significantly,which may be involved in the pathogenesis of viral myocarditis.更多还原
