鹅去氧胆酸衍生物对HepG2裸鼠肝癌移植瘤的影响
Effects of derivatives of chenodeoxychol acid on HepG2 xenografts in nude mice
【摘要】 目的探讨鹅去氧胆酸衍生物对HepG2裸鼠肝癌移植瘤的影响。方法将HepG2细胞注射于SPF级BALB/c裸鼠,形成移植瘤后将瘤体接种于裸鼠,接种成功的小鼠随机分为三组(A组、B组与C组),每组6只裸鼠。A组给予生理盐水腹腔注射,B组给予鹅去氧胆酸衍生物按20 mg/(kg·d)进行腹腔注射,C组给予鹅去氧胆酸衍生物按60 mg/(kg·d)的进行腹腔注射。计算三组的抑瘤率,对肝脏组织进行病理分析与肝癌衍生生长因子(HDGF)表达分析。结果所有裸鼠均接种与造模成功。B组、C组的抑瘤率分别为(38.45±2.15)%和(49.28±3.19)%,C组显著高于B组(P <0.05)。A组肝细胞排列紊乱,肿瘤细胞异型性明显,肝小叶组织正常结构消失,假小叶形成; B组与C组的肝脏病理显著改善(P <0.05),癌症细胞巢形成和细胞坏死显著减少(P <0.05)。三组肝脏组织的HDGF mRNA与蛋白表达水平比较,差异均有统计学意义(P <0.05),B组与C组均显著低于A组(P <0.05)。结论鹅去氧胆酸衍生物可呈剂量依赖性地在HepG2裸鼠肝癌移植瘤中有效发挥抑瘤作用,改善肝脏病理状况,其作用机制可能与抑制HDGF基因与蛋白表达有关。
【Abstract】 Objective To investigate the effects of derivatives of chenodeoxychol acid on HepG2 xenografts in nude mice.Methods The HepG2 cells were injected into SPF BALB/c nude mice,and the tumor were inoculated into nude mice after the formation of the tumor.The successful inoculated mice were randomly divided into three groups(group A,B and C) with 6 nude mice in each group.Group A were given intraperitoneal injection of saline,and group B were given intraperitoneal injection of chenodeoxycholic acid derivatives 20 mg/(kg·d),and group C were given intraperitoneal injection of chenodeoxycholic acid derivatives 60 mg/(kg·d).The inhibition rates of the three groups were calculated,and pathological analysis and HDGF expression analysis of liver tissues were performed.Results The tumor inhibition rate of group B and group C in all nude mice were(38.45 ± 2.15) % and(49.28 ± 3.19) % respectively,with group C higher than group B(P < 0.05).Group A had irregular aligned liver cells and heteromorphic tumor cells with normal structure of the liver lobule disappeared and false lobule formed.The pathological condition of liver in group B and group C were improved significantly,and the cancer nest formation and cell necrosis rate of liver cells decreased significantly(P < 0.05).The difference of HDGF mRNA and protein expression differences among the three groups were statistically significant(P < 0.05),with those of group B and C significantly lower than group A(P < 0.05).Conclusion The application of chenodeoxycholic acid derivatives can effectively suppress tumor and improve pathological condition of liver in HepG2 xenografts in nude mice.The mechanism may be related to the inhibition of HDGF gene and protein expression.
