采用ISSR和SRAP综合分析灰树花的遗传多样性
Genetic Diversity of Grifola frondosa Based on Integrated ISSR and SRAP Analysis
【摘要】 通过菌丝拮抗实验观察灰树花(Grifola frondosa)菌株间拮抗反应及其类型和程度,采用简单序列重复区间(inter-simple sequence repeat, ISSR)和序列相关扩增多态性(sequence-related amplified polymorphism, SRAP)分子标记对灰树花基因组DNA进行PCR扩增,对42株灰树花的遗传多样性进行综合分析。结果表明:拮抗实验将42株灰树花分为17组;筛选出的14条ISSR和11对SRAP引物分别扩增出369条和219条清晰条带,多态性条带平均比率分别为84.01%和81.28%;ISSR和SRAP两种分子标记的综合聚类分析结果较单独分析更准确,当遗传相似系数为0.779时,可将42株灰树花划分为6大类群,表明本研究中选取的42株灰树花菌株的遗传多样性丰富,为杂交育种的亲本选择提供了分子水平的依据。
【Abstract】 Genetic diversity of 42 strains of Grifoloa frondosa was assessed.These strains were initially classified based on their mycelial antagonistic response to each other, and then their ISSR and SRAP molecular markers were amplified from their genomic DNA.The results showed that the 42 strains were divided into 17 groups based on the antagonistic reaction.Using the selected 14 ISSR primers and 11 SRAP primer pairs, 369 ISSR and 219 SRAP bands were obtained with a polymorphic band ratio of 84.01% and 81.28%, respectively.The clustering analysis result based on integration of ISSR and SRAP was more accurate than using either marker individually. The 42 strains were clustered into 6 groups with a genetic similarity coefficient of 0.779, indicating a rich genetic diversity across the G.frondosa strains. These results provided a molecular reference for parental selection in the cross breeding of G.frondosa.
