补充虾青素对人体急性大强度运动后代谢组学的影响

【摘要】 目的:研究服用中等剂量虾青素28d对人体安静状态下血液中小分子代谢物的影响以及持续服用对人体急性大强度运动后机体机能恢复的影响。方法:选取体育学院2016级体教男生8名进行口服虾青素35d(12mg/d)干预。在服用28d后清晨空腹进行一次急性大强度运动:即全力骑功率自行车30s×3/间歇3min,负荷为0. 075kg/kg体重;分别在服用前、服用28d后(运动前)、运动后即刻、运动后1h、运动后1d、运动后1w进行肘静脉采血5mL(全部为空腹采血),对所采血液经离心、去蛋白等处理后进行核磁共振(1HNMR)检测;采用Mest Re Nova、SIMCA-P等软件进行处理与分析并找出差异代谢标志物。结果:补充虾青素28d可使人体安静状态下血浆中乳酸、谷氨酸、谷氨酰胺的水平下降,甘油水平上升;运动后即刻人体血浆中的脂质和甘油、支链氨基酸(亮氨酸、异亮氨酸、缬氨酸)、酮体(β-羟丁酸、乙酰乙酸)、肌酸、肉毒碱、氧化三甲胺、甜菜碱、甘氨酸、葡萄糖水平下降,运动后又在不同时相逐渐恢复。运动后即刻乳酸的水平大幅上升,但在运动1d后恢复至正常值。结论:(1)补充虾青素可影响人体在安静状态下糖的氧化、脂肪代谢、氨基酸代谢等基础代谢过程;(2)急性大强度运动使人体的糖、脂肪、氨基酸代谢等产生一定变化,但在1周内即可恢复。推测其机制可能与补充虾青素加速了运动导致的机体某些代谢物变化的恢复过程有关。

【Abstract】 Objective: To study the effect of small molecule compounds in human blood under resting state with astaxanthin intervention for 28 d and effect of human capability after acute high intensity exercise with continued astaxanthin intervention. Methods: 8 subjects( male students from School of Physical Education) were arranged with astaxanthin intervention( dose of 12 mg/d,taking for 35 d). 28 days later,all subjects underwent acute high intensity exercise in the early morning: 30 s × 3/at three minutes interval,the load is 0. 075 kg/kg body weight.The researchers collected venous blood 5 ml before taking astaxanthin/before exercise( after taking astaxanthin)/immediate post-exercise/1 h post-exercise/1 d post-exercise/1 w post-exercise. Blood samples were underwent1 H-NMR detection after centrifugation,removing protein and other processes. Metabolic profiles were analyzed by Mest ReNova,SIMCA-P and other softwares to find out metabolic markers. Results: The samples are different before and after astaxanthin intervention for 28 d and before and after high-intensity exercise. Supplementation of astaxanthin for 28 d decreased levels of lactic acid,glutamate and glutamine in plasma under resting state,but increased the level of glycerol under resting state. Before and after high-intensity exercise,with the astaxanthin intervention,we found out some metabolic markers in plasma: lipids/BCAAs/keto-bodies/lactate/creatine/carnitine/trimethylamine oxide/betaine/glycine/glucose. All the levels of markers decreaseed except lactate,which was increased. All the levels of markers were received in 1 w,and most of tgen went back to levels under resting state about 1 day. Conclusions:( 1) Supplementation of astaxanthin for 28 d affects glucose metabolism,lipid metabolism and other basic physiological processes.( 2) After acute high intensity exercise,some changes happen on the body’s energy metabolism which involves glucose metabolism,lipids metabolism and protein metabolism,but they can be restored within 1 week. Suggesting that astaxanthin supplementation may accelerate the recovery process of certain metabolites’ changes caused by exercise.
