采用荧光显微技术对枣疯病病原进行鉴定

【摘要】 采集越冬期条枣品种枣疯病枝条对其进行水培,以正常枝条水培为对照,采用荧光显微技术(DAPI)对水培枝条的幼茎进行对比诊断研究。结果表明,越冬期枝条水培采集最佳时间为1月份,水培适宜溶液为1%霍兰格营养液,试材需固定在5%戊二醛溶液中放置于4℃冰箱2 h以上、1.0μg/m L DAPI染色剂中染色20 min,便可快速、便捷、定量定性判断枣疯病病原。

【Abstract】 In this paper, the shoot of Tiaozao variety jujube witches’ broom was collected for hydroponics during overwinter period with normal branches as control, and the young stem of the hydroponics branch was diagnosed by DAPI. The results showed that the best time of hydroponic branches was January, the best hydroponic solution was 1% Hollander nutient solution, the test materials were fixed5% glutaraldehyde in 0.1 mol/L phosphate buffer(pH=7.0)in 4 ℃ refrigerator for over 2 h. They were dyed at least 20 minutes in1.0 μg/m L DAPI, phytoplasma of jujube witches’ broom disease could be judged speedily, conveniently, qualitified and qualitatively.
