对甘薯快繁体系的优化

【摘要】 试验以MS作为基本培养基,设置空白对照,同时添加不同浓度的外源激素(6-BA、NAA、IAA),研究其对不同品种甘薯的不同部位快速繁殖的影响,以此来优化甘薯脱毒苗的快繁体系。结果表明,在成苗时间、生根时间、成苗率等方面,秦薯5号在MS+0.2 mg/L IAA培养基上接种的双叶茎段效果最为突出;红东在MS+0.4 mg/L NAA培养基上接种的双叶茎段效果最好;但从实际生产的角度来讲,还是单叶茎段的增殖效果最好,所以对秦薯5号使用MS+0.2 mg/L IAA培养基接种单叶茎段,对红东使用MS+0.2 mg/L NAA培养基接种单叶茎段;6-BA会抑制根的形成,但是对芽的分化却有促进作用,而且随着6-BA浓度的增长,其对根的抑制效果增强,但对芽的促进作用渐渐消失。研究明确了单一激素在甘薯快繁整体和各阶段的作用,并且选择出了最适合的甘薯快繁切取部位。

【Abstract】 To optimize the rapid propagation system of sweet potato virus-free seedlings, MS was used as the basic medium in this experiment, and a blank control was set. At the same time, different concentrations of exogenous hormones(6-BA, NAA, IAA)were added to treat different parts of different varieties of sweet potato. The results showed that in terms of seedling time, rooting time and seedling rate, the effect of Qinshu 5 on the double-leaf stems inoculated on MS+0.2 mg/L IAA medium was the most prominent,Hongdong in MS+0.4 mg/L NAA medium were the most excellent. However, from the point of view of actual production, the single-leaf stem segment had the best proliferation effect. Therefore, Qinshu 5 was inoculated with single-leaf stem segments using MS+0.2 mg/L IAA medium, MS +0.2 mg/L NAA medium was used for Hongdong. 6-BA inhibited the formation of roots, but promoted the differentiation of buds. With the increase of 6-BA concentration, the inhibitory effect of roots increased, and the promotion of buds gradually disappeared. This study clarified the role of single hormone in the whole and various stages of sweet potato rapid propagation,and selected the most suitable rapid propagation site.
