结缕草高效再生体系的建立

【摘要】 为了获得结缕草的转基因再生植株,从而利用转基因技术改造结缕草,建立草坪草的遗传转化体系,研究了不同培养基、激素浓度及光照条件对结缕草愈伤组织诱导、分化和植株再生的影响,建立了结缕草再生体系。结果表明,在愈伤组织诱导阶段,不同培养基(MS培养基和NB培养基)在同一激素质量浓度下,愈伤组织诱导率差别不大;而同一培养基不同激素浓度间愈伤组织诱导率存在一定差别,以添加2 mg/L的2,4-D处理诱导率较高;光培养和暗培养条件下愈伤组织诱导率差别不大。在愈伤组织的分化阶段,MS培养基远远优于NB培养基,MS培养基上愈伤组织有60%~70%能产生胚性愈伤并得到再生植株,而NB培养基上愈伤组织全部变成绿色的硬块状,并伴随许多毛状根生成,最终不能得到再生植株;暗培养条件下愈伤组织的分化率(55%)和植株再生率(47%)大大高于光培养(10%,7%)。

【Abstract】 To obtain transgenic regenerated plants of Zoysia japonica, Zoysia japonica can be modified by using transgenic technology, the genetic transformation system of Zoysia japonica was established, the effects of different media, hormone concentration and light conditions on callus induction, differentiation and plant regeneration of Zoysia japonica were studied, the regeneration system of Zoysia japonica was established. The results showed that at the stage of callus induction, there was little difference in callus induction rate in different media(MS medium and NB medium). There were some differences in callus induction rates between different hormone concentrations in the same medium, the induction rate of 2, 4-D treatment with 2 mg/L addition was higher. There was little difference in callus induction rate between light culture and dark culture. At the stage of callus differentiation, MS medium was far superior to NB medium, 60%-70% of callus on MS medium could produce embryogenic callus and regenerate plants. The callus on NB medium turned green and hard, and it was accompanied by the formation of many hairy roots, eventually no regenerated plants could be obtained. The differentiation rate of callus(55%)and plant regeneration rate(47%)under dark culture conditions were much higher than those under light culture conditions(10%, 7%).
