柱后补偿电雾式液相色谱仪测定黄芪甲苷含量
Determination of Astragaloside A by Post-Column Compensation Electrospray Liquid Chromatography
【摘要】 本文通过对色谱柱后补偿溶剂的流速、柱温箱温度及电雾式检测器的雾化温度进行优化,确定出采用电雾式检测器测定黄芪甲苷含量的最佳条件。结果表明:在柱后补偿中最佳的测定条件为乙腈流速0.4 mL/min,柱温30℃,雾化器温度30℃。优化后的检测方法重现性好、灵敏度高,操作简单,适用于采用电雾式检测器测定黄芪甲苷的含量。
【Abstract】 The optimum separation conditions for determination of astragaloside A were determined by optimizing the flow rate of compensating solvent after column, column temperature and atomization temperature of electrospray detector. The results show that the optimum conditions for post-column compensation are acetonitrile flow rate of 0.4 mL/min, column temperature of 30 ℃ and atomization temperature of 30 ℃. The method has good reproducibility, high sensitivity and simple operation. It is suitable for the determination of astragaloside A in Astragalus membranaceus.
