甘扎嘎日果总酚纯化工艺及抗氧化活性

【摘要】 目的:筛选纯化甘扎嘎日果总酚的大孔树脂,确定其纯化工艺,并评价甘扎嘎日果总酚的体外抗氧化活性。方法:以吸附-解吸率为指标,通过对比研究AB-8、D101、HPD100、HPD600、HPD826、NKA-9、ADS-17共7种大孔树脂的纯化效果,筛选出纯化甘扎嘎日总酚的树脂,采用单因素试验确定大孔树脂纯化甘扎嘎日果总酚的工艺条件,利用自由基消除试验和总还原能力测定对甘扎嘎日果总酚物质的抗氧化活性进行评价。结果:ADS-17型大孔树脂纯化甘扎嘎日果总酚的效果更佳,得率可达62.51 mg/g,为纯化前的17.5倍,纯化工艺参数为:上样液浓度1.5 mg/mL、流速为1 mL/min、树脂径高比为1:11、上样量为110 mL,140 mL 20%乙醇作为除杂剂,140 mL 80%乙醇为洗脱剂。DPPH自由基消除试验和总还原能力测定结果显示,甘扎嘎日果总酚纯化物抗氧化活性优于其提取物,但甘扎嘎日果总酚的抗氧化活性略弱于阳性对照物Vc。结论:研究所用的纯化方案可实现甘扎日果总酚的富集纯化,且纯化得到的甘扎嘎日果总酚具有较强的抗氧化活性。

【Abstract】 Objective: The resin for the purification of total phenol in Kandra Kari fruit was screened and the purification process was determined, and its antioxidant activity were evaluated in vitro. Methods: The best resin for purifying total phenol was screened out from seven resins such as AB-8, D101, HPD100, HPD600, HPD826, NKA-9 and ADS-17 by adsorption-resolution rate, the purification process for total phenol in Kandra Kari fruit was determined by the single-factor test, and the antioxidant activity of total phenol was evaluated by free radical elimination test and total reduction ability. Results: The ADS-17 is the best resin for purifying total phenol in Kandra Kari fruit, and the content of total phenol is 62.51%, which is 17.5 times than the content of extract. The process parameters were as follows: The concentration was 1.5 mg/mL, leaching rate of elutriation agent was 1.0 mL/min, the resin diameter to height ratio was 1:11, the dosage of sample was 110 mL, 140 mL of 20% ethanol was used to remove impurity, and 140 mL of 80% ethanol was used as its eluting agent. The results of antioxidant experiments showed that the total phenol in Kandra Kari had strong antioxidant activity, and its activity was stronger than that of extract, but it was weaker than that of the Vc. Conclusion: The process could be used for the purification and enrichment of total phenol from Kandra Kari fruit, and the total phenol has better antioxidant activity.
