HPLC法同时测定蜂蜜中9种防腐剂、甜味剂和色素

【摘要】 建立了蜂蜜中苯甲酸、山梨酸、糖精钠、安赛蜜、柠檬黄、日落黄、胭脂红、苋菜红和脱氢乙酸的高效液相色谱测定方法。样品采用甲醇-水溶液(1︰5, V/V)提取,色谱柱为Eclipse XDB-C18 (250 mm×4.6 mm, 5μm),以0.02 mol/L乙酸铵溶液和甲醇为流动相梯度洗脱,检测波长为230 nm,检测的线性范围为10~100μg/m L。结果表明, 9种物质在线性范围内具有良好的线性关系,相关系数≥0.999 2。9种物质的最低检出限为0.08~0.34 mg/kg。在低、中、高(10, 40和80μg/m L) 3个加标浓度下,方法的加标回收率为96.32%~105.78%,相对标准偏差(RSD)为1.69%~3.67%(N=6)。该方法灵敏、简单、快速、准确可靠,适用于大批量蜂蜜中常见添加剂的检测。

【Abstract】 A method was developed to separate and detect the benzoic acid, sorbic acid, saccharine sodium, anselmine, lemon yellow, sunset yellow, rouge red, amaranth red and dehydroacetic acid in honey by high performance liquid chromatography.The honey samples were extracted with methanol-water solution(1︰5, V/V). The chromatographic column was Welch Xtimate C18 column(250 mm×4.6 mm, 5 μm), using methanol and(0.02 mol/L) acetic acid ammonium system for gradient elution.The detection wavelength was set at 230 nm, and the linear range of detection was 10-100 μg/mL. The results showed that the nine substances had good linear relations in the linear range, and the correlation coefficients were more than 0.999 2. The minimum detection limit was 0.08-0.34 mg/kg. The recoveries of nine compounds at three spiked levels(10, 40 and 80 μg/mL) were in the range of 96.32% to 105.78%, and the relative standard deviations(RSDs) ranged from 1.69% to 3.67%(N=6). The method was simple, rapid and sensitive, and could be used for the detection of common additives in honey.
