改良推注方法制备脂肪移植SVF胶的实验研究

【摘要】 目的探讨改良推注法与常规推注法制备基质血管成分(stromal vascular fraction, SVF)胶的效果。方法分别采用改良推注法和常规推注法从抽吸获取的脂肪组织中分离制备SVF胶,并对所获取的SVF胶进行细胞计数、增殖率、细胞成活率、细胞碎片率等生物学特性进行检测。结果改良推注法获取的SVF胶的细胞成活率较高(P<0.05),细胞凋亡、细胞碎片率显著低于常规推注法(P<0.05)。结论采用改良推注法制备脂肪移植SVF胶更加快捷、方便,并且可提高移植细胞的成活率、降低细胞碎片率,在疾病治疗及创伤修复中有较好的应用前景。

【Abstract】 Objective To explore the efficacy of the modified non-enzymatic isolation method and the conventional method in the preparation of stromal vascular fraction(SVF) gel. Methods The SVF gel was prepared through this conventional method as well as the modified non-enzymatic isolation method from the obtained adipose tissue. The biological characteristics of SVF gel such as cell count,proliferation rate, cell viability, and cell debris were measured. Results The cell viability of SVF gel extracted through the modified non-enzymatic isolation method was higher(P<0.05)and apoptosis and cell debris were lower than that of conventional method(P<0.05).Conclusion The modified non-enzymatic isolation method was rapid and convenient for the preparation of SVF gel. This method increased cell viability, decreased cell debris, and had broader clinical application prospects in the repair of trauma and treatment of disease compared to the conventional method.
