补髓生血颗粒对慢性髓劳病IL-21和IL-21mRNA的影响

【摘要】 目的通过观察补髓生血颗粒对慢性髓劳病IL-21和IL-21mRNA的影响,分析IL-21和IL-21mRNA与慢性髓劳病发病的相关性,进一步探讨补髓生血颗粒调整免疫失衡的疗效机制。方法选取血液病科2013年8月至2015年3月期间门诊和住院的慢性髓劳病患者30例为试验组,口服补髓生血颗粒治疗,以同期10名健康志愿者为正常对照组。试验组应用ELISA法检测治疗前后IL-21表达变化,RT-PCR法检测治疗前后IL-21mRNA表达的变化。结果试验组慢性髓劳病IL-21和IL-21mRNA的表达高于正常对照组(P <0. 05);试验组治疗后IL-21和IL-21mRNA的表达较治疗前下降(P <0. 05),差异有统计学意义。结论 IL-21和IL-21mRNA在慢性髓劳病中的异常表达,提示可能参与了慢性髓劳病的免疫发病过程;补髓生血颗粒通过下调IL-21和IL-21mRNA的表达,以调整异常的免疫反应,进而恢复骨髓的造血功能。

【Abstract】 Objective To analyze the correlation of IL-21 and IL-21 mRNA with chronic aplastic anemia(CAA) through observing the impacts of busui shengxue granules on IL-21 and IL-21 mRNA before and after treatment in the CAA patients so as to further explore the mechanism of busui shengxue granules on regulating immune imbalance.Methods From August 2013 to March 2015,30 outpatients or inpatients with CAA were collected from the Department of Hematology in a trial group,and they were treated with the oral administration of busui shengxue granules. Simultaneously,10 healthy volunteers were collected in a normal control group. Before and after treatment,ELISA was adopted to determined the expressions of IL-21 and RT-PCR to determine the expressions of IL-21 mRNA in the trial group. Results In the trial group,the expressions of IL-21 and IL-21 mRN in CAA patients in the trial group were higher than the normal control group(P < 0. 05). After treatment,the expressions of IL-21 and IL-21 mRNA in the trial group were reduced as compared with those before treatment(P < 0. 05),indicating the statistical significance. Conclusion The abnormal expressions of IL-21 and IL-21 mRNA in CAA suggests that they may be participated in the immune pathogenesis of CAA.Busui shengxue granules regulates the abnormal immune reaction and recover hematopoietic function of bone marrow through down-regulating the expressions of IL-21 and IL-21 mRNA.
