【摘要】 目的探讨中药金银花对病毒性心肌炎小鼠Caspase-3、NF-κB表达的影响。方法将80只雄性BALB/c小鼠随机分为4组,每组20只,分别为正常对照组、模型组、金银花低剂量组和金银花高剂量组。模型组和金银花高、低剂量组小鼠均腹腔注射柯萨奇病毒B3(CVB3),建立病毒性心肌炎模型。金银花高、低剂量组于注射病毒后30 min开始灌服金银花药液,每日1次,共14 d;模型组以0.9%NaCl溶液灌胃;正常对照组用不含病毒的等量Eagle液腹腔注射,等量的0.9%NaCl溶液灌胃。各组分别在14 d随机抽取15只小鼠处死,HE染色观察心肌病理改变,免疫组化检测心肌组织中Caspase-3、NF-κB的表达。结果模型组的Caspase-3、NF-κB表达水平均明显高于正常对照组,金银花高、低剂量组的Caspase-3、NF-κB表达较模型组均明显减轻;金银花高剂量组的Caspase-3、NF-κB表达水平较金银花低剂量组减轻。结论金银花可明显抑制病毒性心肌炎小鼠Caspase-3、NF-κB的表达,为临床应用金银花治疗病毒性心肌炎提供了实验依据。更多还原

【Abstract】 Objective To investigate the effect of honeysuckle on the expression of Caspase-3 and NF-κB in mice with viral myocarditis. Methods A total of 80 male BALB/c mice were divided into 4 groups randomly with 20 mice in each group:normal control group,model group,honeysuckle low-dose group and honeysuckle high-dose group. Coxsackievirus B3(CVB3) was injected intraperitoneally into mice in model group and both dose groups to establish viral myocarditis model. The low and high dose groups were given honeysuckle liquid 30 minutes after virus injection, once a day for 14 days; the model group was given saline by gastric lavage; the control group was given intraperitoneal injection of Equivalent Eagle liquid without virus and normal saline by gastric lavage. Totally 15 mice were randomly selected and executed from each group on the 14 th day. The pathological changes of myocardium were observed by HE staining, and the expression of Caspase-3, NF-κB in myocardial tissue was detected by immunohistochemistry. Results The expression of Caspase-3 and NF-κB in the model group was significantly higher than that in the control group. The expression of Caspase-3 and NF-κB in the two dose groups was significantly lower than that in the model group. The expression of Caspase-3 and NF-κB in the honeysuckle high-dose group was significantly lower than that in the honeysuckle low-dose group. Conclusion Honeysuckle can significantly inhibit the expression of Caspase-3 and NF-κB in mice with viral myocarditis, which provides the experimental basis for clinical application of honeysuckle in the treatment of viral myocarditis.更多还原
